RINGKASAN
Derajat Hubungan Tingkat Keparahan Infeksi Karat Daun Kedelai dengan
Produksi pada Generasi Segregasi Kelima Genotipe UNEJ-1 dan UNEJ-2.

Kebutuhan kedelai nasional pada saat ini belum terpenuhi sehingga Indonesia masih impor, karena saat ini komoditas kedelai tidak hanya diposisikan
sebagai bahan pangan dan bahan baku industri pangan tetapi juga ditempatkan
sebagai bahan makanan sehat dan bahan baku industri non-pangan. Penyebab
utama rendahnya produksi kedelai di Indonesia ialah masalah gangguan hama dan
penyakit tanaman. Penyakit yang sering merusak tanaman kedelai ialah karat
daun kedelai, Phakopsora pachyrhizi Syd. Salah satu upaya untuk mengatasi
penurunan produksi kedelai akibat karat daun kedelai yaitu dengan penggunaan
varietas tahan yang memiliki keunggulan produksi tinggi.
Genotipe kedelai UNEJ-1 dan UNEJ-2 yang dilaporkan sebagai genotipe
dengan keunggulan produksi tinggi dan agak tahan karat daun kedelai, saat ini
sifat keunggulannya dikembangkan dengan mengintroduksikan sifat ukuran biji
besar dari kedelai Edamame (Ryokoh-75) melalui uji silang balik. Pada setiap
tahap hasil uji silang balik dalam proses perbaikan genotipe tersebut, respon
genotipe UNEJ-1 dan UNEJ-2 terhadap infeksi patogen karat daun kedelai masih
perlu diuji. Pada penelitian ini, pengujian dilakukan terhadap hasil uji silang balik
pada generasi segregasi kelima genotipe UNEJ-1 dan UNEJ-2. Pengujian
dimaksudkan untuk mengetahui derajad hubungan tingkat keparahan infeksi karat
daun kedelai pada dua genotipe tersebut dengan produksi, dan menilai sejauh
mana tingkat keparahan infeksi karat daun kedelai tersebut berdampak terhadap
penurunan produksi.
Benih kedelai genotipe UNEJ-1 dan UNEJ-2, ditanam pada petak percobaan berukuran 0,4 m x 4,5 m (berisi 160 tanaman) dengan jarak lubang tanam
40 cm x 15 cm dan setiap lubang tanaman digunakan dua benih. Pengujian
dilaksanakan di kebun percobaan BALITKABI Muneng Probolinggo pada musim

tanam MK-1. Infeksi karat daun kedelai terjadi secara alami, dan tingkat
keparahan penyakit karat daun kedelai pada setiap genotipe diamati pada tanaman
umur 60, 70, dan 80 hari setelah tanam (HST). Keparahan penyakit diukur dengan
menentukan intensitas penyakit berdasarkan katagori kepadatan bercak karat
dengan skala 1-4, menggunakan rumus: IP = [∑(n x v)/(N x V) ] X 100%
(IP = intensitas penyakit, n = jumlah daun pada setiap katagori, v = nilai skala
kepadatan bercak karat 1, 2,.., dan 4; N = jumlah daun yang diamati, V = nilai
skala tertinggi). Skala kepadatan bercak karat pada daun yaitu 1 = tidak ada
bercak karat/cm², 2 = 1-8 bercak karat/cm², 3 = 9-16 bercak karat/cm², dan
4 = lebih 16 bercak karat/cm².
Pengukuran IP dilakukan pada daun-daun dari 1/3 tanaman bagian paling
bawah, 1/3 tanaman bagian tengah, dan 1/3 tanaman bagian paling atas pada
setiap tanaman contoh. Hubungan tingkat keparahan penyakit karat daun kedelai
dengan produksi diamati pada setiap genotipe yang diuji dengan mengukur
komponen produksi yang meliputi jumlah polong (hampa dan bernas), jumlah biji
per tanaman, berat biji per tanaman, dan berat 100 biji. Percobaan disusun
berdasarkan rancangan acak kelompok (RAK) subsampling terdiri atas dua
perlakuan (genotipe), tiga ulangan dan 10 anak contoh. Perbedaan antar perlakuan
dianalisis dengan uji DMRT pada taraf kepercayaan 5%.
Hasil yang diperoleh terdapat perbedaan tingkat keparahan penyakit antara
genotipe UNEJ-1 dengan genotipe UNEJ-2. Tingkat keparahan penyakit pada
genotipe UNEJ-1 menunjukkan nilai IP lebih tinggi dibandingkan dengan
genotipe UNEJ-2, namun tingkat keparahan kedua genotipe tersebut masih
dikatagorikan sedang. Ada korelasi antara tingkat keparahan penyakit karat daun
kedelai dengan beberapa komponen produksi, dan terdapat korelasi negatif
khususnya pada berat biji per tanaman. Genotipe UNEJ-1 juga menunjukkan
penurunan berat 100 biji lebih besar dibandingkan genotipe UNEJ-2, tetapi
UNEJ-1 masih memiliki kelebihan dengan ukuran biji yang lebih besar.

RINGKASAN
Derajat Hubungan Tingkat Keparahan Infeksi Karat Daun Kedelai dengan
Produksi pada Generasi Segregasi Kelima Genotipe UNEJ-1 dan UNEJ-2.

Universitas Jember.
Kebutuhan kedelai nasional pada saat ini belum terpenuhi sehingga Indonesia masih impor, karena saat ini komoditas kedelai tidak hanya diposisikan
sebagai bahan pangan dan bahan baku industri pangan tetapi juga ditempatkan
sebagai bahan makanan sehat dan bahan baku industri non-pangan. Penyebab
utama rendahnya produksi kedelai di Indonesia ialah masalah gangguan hama dan
penyakit tanaman. Penyakit yang sering merusak tanaman kedelai ialah karat
daun kedelai, Phakopsora pachyrhizi Syd. Salah satu upaya untuk mengatasi
penurunan produksi kedelai akibat karat daun kedelai yaitu dengan penggunaan
varietas tahan yang memiliki keunggulan produksi tinggi.
Genotipe kedelai UNEJ-1 dan UNEJ-2 yang dilaporkan sebagai genotipe
dengan keunggulan produksi tinggi dan agak tahan karat daun kedelai, saat ini
sifat keunggulannya dikembangkan dengan mengintroduksikan sifat ukuran biji
besar dari kedelai Edamame (Ryokoh-75) melalui uji silang balik. Pada setiap
tahap hasil uji silang balik dalam proses perbaikan genotipe tersebut, respon
genotipe UNEJ-1 dan UNEJ-2 terhadap infeksi patogen karat daun kedelai masih
perlu diuji. Pada penelitian ini, pengujian dilakukan terhadap hasil uji silang balik
pada generasi segregasi kelima genotipe UNEJ-1 dan UNEJ-2. Pengujian
dimaksudkan untuk mengetahui derajad hubungan tingkat keparahan infeksi karat
daun kedelai pada dua genotipe tersebut dengan produksi, dan menilai sejauh
mana tingkat keparahan infeksi karat daun kedelai tersebut berdampak terhadap
penurunan produksi.
Benih kedelai genotipe UNEJ-1 dan UNEJ-2, ditanam pada petak percobaan berukuran 0,4 m x 4,5 m (berisi 160 tanaman) dengan jarak lubang tanam
40 cm x 15 cm dan setiap lubang tanaman digunakan dua benih. Pengujian
dilaksanakan di kebun percobaan BALITKABI Muneng Probolinggo pada musim

tanam MK-1. Infeksi karat daun kedelai terjadi secara alami, dan tingkat
keparahan penyakit karat daun kedelai pada setiap genotipe diamati pada tanaman
umur 60, 70, dan 80 hari setelah tanam (HST). Keparahan penyakit diukur dengan
menentukan intensitas penyakit berdasarkan katagori kepadatan bercak karat
dengan skala 1-4, menggunakan rumus: IP = [∑(n x v)/(N x V) ] X 100%
(IP = intensitas penyakit, n = jumlah daun pada setiap katagori, v = nilai skala
kepadatan bercak karat 1, 2,.., dan 4; N = jumlah daun yang diamati, V = nilai
skala tertinggi). Skala kepadatan bercak karat pada daun yaitu 1 = tidak ada
bercak karat/cm², 2 = 1-8 bercak karat/cm², 3 = 9-16 bercak karat/cm², dan
4 = lebih 16 bercak karat/cm².
Pengukuran IP dilakukan pada daun-daun dari 1/3 tanaman bagian paling
bawah, 1/3 tanaman bagian tengah, dan 1/3 tanaman bagian paling atas pada
setiap tanaman contoh. Hubungan tingkat keparahan penyakit karat daun kedelai
dengan produksi diamati pada setiap genotipe yang diuji dengan mengukur
komponen produksi yang meliputi jumlah polong (hampa dan bernas), jumlah biji
per tanaman, berat biji per tanaman, dan berat 100 biji. Percobaan disusun
berdasarkan rancangan acak kelompok (RAK) subsampling terdiri atas dua
perlakuan (genotipe), tiga ulangan dan 10 anak contoh. Perbedaan antar perlakuan
dianalisis dengan uji DMRT pada taraf kepercayaan 5%.
Hasil yang diperoleh terdapat perbedaan tingkat keparahan penyakit antara
genotipe UNEJ-1 dengan genotipe UNEJ-2. Tingkat keparahan penyakit pada
genotipe UNEJ-1 menunjukkan nilai IP lebih tinggi dibandingkan dengan
genotipe UNEJ-2, namun tingkat keparahan kedua genotipe tersebut masih
dikatagorikan sedang. Ada korelasi antara tingkat keparahan penyakit karat daun
kedelai dengan beberapa komponen produksi, dan terdapat korelasi negatif
khususnya pada berat biji per tanaman. Genotipe UNEJ-1 juga menunjukkan
penurunan berat 100 biji lebih besar dibandingkan genotipe UNEJ-2, tetapi
UNEJ-1 masih memiliki kelebihan dengan ukuran biji yang lebih besar.
