Abstract
This study was aimed

to analyze the effect of patchouli oil from Nilam on the expression of calcium

sensing receptor (CaSR) and Ki-67 in breast cancer cells. This research is in vitro against breast cancer
cells MCF-7. The study group includes a group of control cells {MCF-7 cells without any treatment} and

a group of MCF-7 cells were incubated with Patchouli at doses 156, 48 pM , 312, 96 UM , and
pM with duration of incubation was 12 and 24 hours. Analysis CaSR expression and Ki-67 was done by
confocal laser scanning microscpe. For the 12-hour incubation, there were no significant differences in
the expression of CaSR and Ki-67 among the study group (D 0.05). For the 24-hour incubation, CaSR

expression and Ki-67 were decreased significantly in all dose Patchouli compared with the control

Untuk inkubasi 24 jam, ekspresi CaSR dan Ki-6Tmenurun secara bermakna pada semua dosis Patchouli
dibqndingkan kelompok kontrol {P < 0.05). Dosis optimum penurunan CaSR dan Kl-67 adalah dosis
312,96pM. Disimpulkan bahwa Patchoulimemicu penghambatan proliferasisel kanker payudara MCF7 pada inkubasi 24 jam dengan dosis optimum pada dosis kedua.

ABSTRACT
This study was aimed to analyze the effect of patchouli oil from Nilam on the expression of

For the 24-hour incubation, CaSR and Ki-67 expressions significantly decreased in all
Patchouli dosages than the control group (P < 0 05). The optimal dosage for reduction of
CaSR and Kl-67 is 312.96 ptr/ (Gambaran sel IICF sama dengan sel ttilCF pada figure 1)

Calcium sensing receptor (CaSR) is found in normal breast tissues and malignant breast
tissues [7]. Caleium sensing receptor is oncogenic in breast 12,4,51.|n this research, for the
24-hour incubation, CaSR expression is significantly lower in all three Patchouli alcohol
dosages than the control group (P < 0.05). Patchouli administration at the dosage of 312.96

pM may suppress the expression of CaSR equal to the expression in control group (P >
0.05). This indicates that Patchouli alcoholtriggers down-regulation of CaSR expression as

an oncogene in breast cancer cell [/lCF-7. According to the researchers' knowledge, this is
the first research to evaluate the effects gl Patchouli alcohol on CaSR as the oncogene in
breast cancer. Analogous to colorectal cancer, the mechanism of CaSR down regulation is

mechanism is through NF-KB when CaSR genes have functional elements of kappa b [19211.

The researchers hypothesize that the mechanism of down regulation of CaSR is least

through aberrant DNA methylation, histone deacetylation, and NF-KB. Then this down
regulation is followed by the decrease in cell proliferation, characterized by the decrease in

Ki-67 expression

in MCF-7 cells. This suggests lhal Patchouli a/cohol has

anticancer

property by inhibiting the proliferation of l\4CF-7 cells that at least is through CaSR downregulation mechanism. This study is consistent with previous findings, saying that Patchoull

a/corol is able to inhibit the groMh of NICF-7 cells

It is concluded that Patchouli alcoholltiggets the inhibition of the NICF-7 breast cancer cells
proliferation in 24-hour incubation with the optimum dosage of 312.96 plvl. Thus, we are sure
that Patchouli alcohol can be a pharmacological agent for breast cancer treatment.
