Media merupakan salah satu faktor penting dalam mendukung pertumbuhan
anggrek Cattleya ‘Blc. Mount Hood Mary’ secara in vitro. Penambahan bahan
organik pada media kultur telah diketahui dapat meningkatkan pertumbuhan
anggrek Cattleya. Tujuan dari percobaan ini adalah untuk mendapatkan pengaruh
terbaik dari kombinasi konsentrasi ekstrak wortel (EW) dan air kelapa (AK) pada
multiplikasi dan pertumbuhan tunas Cattleya ‘Blc. Mount Hood Mary’. Percobaan
ini dilaksanakan di Laboratorium Kultur Jaringan Teknologi Benih,
Fakultas Pertanian, Universitas Padjadjaran pada bulan Februari sampai April
2012. Eksplan yang digunakan adalah tunas mikro yang berasal dari
perkecambahan biji Cattleya ‘Blc. Mount Hood Mary’. Rancangan yang
digunakan adalah Rancangan Acak Lengkap dengan delapan perlakuan, dan
diulang empat kali. Media dasar Murashige dan Skoog digunakan sebagai media
dasar. Delapan perlakuan diaplikasikan pada media dasar yang terdiri dari
kombinasi konsentrasi EW dan AK. EW terdiri dari empat konsentrasi yaitu 25,
50, 100, dan 150 ml L-1 dan CW terdiri dari dua konsentrasi yaitu 200 ml L-1 dan
kontrol (tanpa air kelapa). Hasil peneltian menunjukkan bahwa medium yang
mengandung 50 ml L-1 EW + 200 ml L-1 AK; 100 ml L-1 EW + 200 ml L-1 AK;
100 ml L-1 EW + 0 ml L-1 menunjukkan rata-rata pertambahan jumlah tunas dan
daun yang tinggi. Medium yang dilengkapi dengan 50 ml L-1 EW + 200 ml L-1
AK; 100 ml L-1 EW + 0 ml L-1 AK memberikan pengaruh yang baik terhadap
tinggi plantlet. Medium yang mengandung 100 ml L-1 EW + 200 ml L-1 AK
memberikan pengaruh terbaik untuk bobot segar yaitu 50,1625 mg.

Media is one of the important factor to support the growth of Cattleya ‘Blc.
Mount Hood Mary’ in vitro. Organic material addition to culture media has been
known to promoted the growth of Cattleya. The aim of this experiment was to
obtain the best influence of carrot extract (CE) and coconut water (CW)
concentration to the shoot multiplication and growth of Cattleya ‘Blc. Mount
Hood Mary’. This research was carried out at Tissue Culture Laboratory of Seed
Technology, Faculty of Agriculture, Universitas Padjadjaran during February until
April 2012. Shoot derived seed of Cattleya ‘Blc. Mount Hood Mary’ explant was
used in this experiment. A Completely Randomized Design was used in this
experiment with eight treatments and replicated four times. Murashige and Skoog
media was used as basic media. Eight treatments was applied in this basic media
consisted of combination of CE and CW. CE consisted of four consentrations
namely 25, 50, 100, and 150 ml L-1 and CW consisted of two consentrations
namely 200 ml L-1 and control (without CW). Result shown that medium
supplemented with 50 ml L-1 CE + 200 ml L-1 CW; 100 ml L-1 CE + 200 ml L-1
CW; 100 ml L-1 CE + 0 ml L-1 CW shown the high average number of shoots and
leaves increment. Medium supplemented with 50 ml L-1 CE + 200 ml L-1 CW;
100 ml L-1 CE + 0 ml L-1 CW gave a good effect for plantet height. Medium
supplemented with 100 ml L-1 CE + 200 ml L-1 CW gave a good effect for fresh
weight that was 50.1625 mg.
