Transmisi Transgen (PhGH) dan Performa Pertumbuhan Ikan Lele (C. gariepinus) Transgenik F3 di Balai Penelitian Pemuliaan Ikan Sukamandi Subang, Jawa Barat

ABSTRAK
SULTAN AKBAR HABIBULLAH. Transmisi Transgen (PhGH) dan Performa
Pertumbuhan Ikan Lele (C. gariepinus) Transgenik F3 di Balai Penelitian
Pemuliaan Ikan Sukamandi Subang, Jawa Barat. Dibimbing oleh ZULKIFLI
NASUTION, YUNASFI dan HURIA MARNIS.
Perkembangan produksi perikanan dibidang akuakultur membutuhkan
adanya pemenuhan kebutuhan akan induk unggul untuk mempercepat laju
produksi. Teknologi transgenesis dengan introduksi transgen (PhGH) dapat
dilakukan untuk menghasilkan ikan cepat tumbuh dan memiliki performa yang
lebih baik. Deteksi transgen dilakukan dengan menggunakan metode PCR
(Polymerase Chain Reaction) dan elektroforesis menggunakan benih lele
transgenik F3 progeni. Selanjutnya dilakukan pengamatan terhadap performa ikan
melalui pengukuran derajat pembuahan (jumlah telur terbuahi/total telur), derajat
penetasan (jumlah telur menetas/jumlah telur dibuahi), tingkat kelangsungan
hidup (jumlah ikan awal/jumlah ikan akhir), performa pertumbuhan (bobot ratarata transgenik F3/bobot rata-rata non-transgenik) dan efisiensi pakan (laju
pertumbuhan mutlak/konsumsi pakan harian). Hasil menunjukkan bahwa tingkat
transmisi berkisar antara 0-75% dengan rata-rata transmisi pada induk betina
pembawa transgen sebesar 2,5% (15/600) dan 14,4% (75/520) pada induk jantan
pembawa transgen. Ikan lele transgenik F3 memiliki derajat pembuahan 0,37%
lebih kecil dan memiliki derajat penetasan 4% lebih besar dibanding nontransgenik. Efisiensi pakan ikan lele transgenik lebih tinggi dibanding nontransgenik Hal ini menunjukkan ikan lele transgenik memiliki performa yang
lebih baik dibanding dengan non-transgenik.

Developments in the field of aquaculture fish production requires the
fulfillment of the need to accelerate the pace brood production. Transgenesis
technology with the introduction of transgenes (PhGH) can be performed to
produce the fish grow faster and have better performance. Transgene detection
was performed using the PCR method (Polymerase Chain Reaction) and
electrophoresis using transgenic catfish seed F3 progeny. Furthermore, the
observation of the performance of catfish transgenic F3 by measuring the degree
of fertilization (the number of fertilized eggs / total eggs), hatching (the number of
eggs hatched / number of fertilized eggs), the survival rate (the number of early
fish / amount of fish end), the performance of growth (weight Average transgenic

F3 / weight average non-transgenic) and feed efficiency (absolute growth rate /
daily feed intake). Results indicate that the transmission rate ranged from 0-75%
with an average transmission in the female parent carriers of the transgene by
2.5% (15/600) and 14.4% (75/520) in the male carriers of the transgene. F3
transgenic catfish have a degree of fertilization 0.37% smaller and has a hatching
4% higher than the non-transgenic. Transgenic catfish feed efficiency higher than
non-transgenic This shows transgenic catfish have better performance compared
with non-transgenic.
