ABSTRACT
TRI RATNA SULISTIYANI. Isolation and Characterization of Antibiotic
Produced by Actinomycetes from West Timor Island (NTT)Soils. Under the
direction of MANSJUR HAWAB, IWAN SASKIAWAN, and NUNUIC
WIDHYASTUTI.
Actinomycetes is known have potential as source of various bioactive
compounds used in pliarmaceutical industry, agriculture and others purpose. Local
Actinomycetes are capable of producing secondaly metabolites useful antibiotics
was done against Micrococcus luteus, Staphylococcus aureus, Eschericia coli,
BacciEus szlbtilis, Srrcharolnyces cerrevisiae, dan Candida albicans.
This research is divided into isolation of Actinomycetes from west Timor
Island (NTT) and characterization antibiotic activity. The selected crude extract
(A1 6.1) fro111 production medium were characterized for various pH (acid, neutral,
and alkaline) and temperature (30-100 OC). Antibiotics extraction was done by
solvent hexane, chloroform, and ethyl acetate. Analysis of antibiotic substance
was done by thin layered chromatography (TLC) and bioautography.
Sevellfy-four isolates among 74 isolated Actinomyctes are arltibiotics
producer inhibited M. luteus, S. aureus, 3.substillis, E. coli, S. cerrevisiae, and
C. albicans. Antibiotic produced by isolate A16.1 is relatively stabile in extensive
pH and teniperature 30-100 OC. Ethyl acetate is the best solvent to extract
is proved
antibiotic isolate A 16.1. The result fiom TLC with chloroform:metl~aiio~
2 spot and result from bioautography is spot with Rf 0.4 have antirnikrobe
