Pengaruh Ekstrak Etanol Buah Labu Siam (Sechium edule Jacg. Swartz.) Terhadap Kadar Interleukin 6 Pada Mencit Hiperglikemia Yang Diinduksi Streptozotocyn (STZ)

Abstrak
Diabetes mellitus menjadi ancaman global yang bersifat serius dengan
prevalensi yang terus meningkat. Pembuatan hewan model diabetes mellitus
menggunakan Streptozotocin (STZ) akan menyebabkan peningkatan kadar radikal
bebas dan inflamasi pada pankreas sehingga merusak sel-sel β. Ekstrak etanol
buah Labu Siam (Sechium edule,Jacq.Swartz.)merupakan tumbuhan yang
mengandung flavonoid dan salah satu sifatnya antiinflamasi.
Penelitian ini bertujuan untuk mengetahui pengaruh ekstrak etanol buah
Labu Siam terhadap penurunan kadar Gula Darah (KGD), kadar Interleukin 6
(IL-6) dan peningkatan ukuran diameter pankreas pada mencit hiperglikemia
yang diinduksi STZ.
Penelitian ini adalah penelitian eksperimental dengan rancangan post test
randomized controlled group design pada mencit jantan yang dibagi menjadi 4
kelompok yaitu kelompok kontrol negatif, kontrol positif (streptozotocin dosis 60
mg/kgBB), dan 2 kelompok perlakuan (dosis 100 gram/kg BB, dan 200 gram/kg
BB).
Pengukuran kadar IL-6 dilakukan dengan metoda ELISA, kadar gula darah
menggunakan glukometer dan histopatologis pankreas dilakukan dengan
mengamati perubahan ukuran diameter pankreas mencit secara mikroskopis.
Analisa data kadar KGD dan IL-6 menggunakan One-Way ANOVA (α = 0,005).
Hasil penelitian menunjukkan terjadinya penurunan kadar IL-6 walaupun

tidak signifikan bila dibandingkan dengan kontrol positif. Pengamatan preparat
histopatologi pankreas mengalami peningkatan ukuran diameter yang
menunjukkan adanya pelebaran sel β pankreas.
Simpulan dari penelitian ini adalah Ekstrak etanol buah Labu Siam dapat
menurunkan kadar IL-6 serta menghambat kerusakan pankreas pada mencit
hiperglikemia.

Abstract
Diabetes mellitus becomes a global serious health threat with definite
increasing prevalence. Diabetes mellitus animal model was made by induction of
Streptozotocin (STZ) that causes increasing of free radical level and inflammation
in pancreas organ that destroys β cell. Squashfruitethanol extract of Chayotte
(Sechium edule)is a plantcontainsflavonoids one of characterisanti-inflammatory.
The objectives of this study are determining the effectof ethanol extract
ofthe fruitSquashChayotte (Sechium edule), decreasingblood sugarlevels,
Interleukin 6 (IL-6) level and increasingthe size ofthe diameter ofthe pancreas
inSTZ-induced hyperglycemicmice model.
This research is experimental research uses Posttest Only Group Design

Method by Completely Randomized Design in male mice and divided into 4
groups; negative control group, positive control group is treated with
streptozotocin at a dose of 60 mg/kg weight, and 2 groups of therapy (dosage 100
gram/kg weight, 200 gram/kg weight).
IL-6 level measurement with ELISA method, blood sugar and
histopathological pancreas done by observing microscopis the ateration of the size
of diameter pancreas mice. Analysis of blood sugar level, IL-6 level uses One
Way ANOVA(α = 0,005) and observe the increase diameter of pancreas using
microscope .
The results showed a decrease in the levels of IL-6, although it’s not
significant when compared with the positive control. Histopathological
observation of pancreatic preparations increased diameter which showed widening
of pancreatic β cells.
In conclusion, the decreases IL-6 level and islet of Langerhans damage in
hyperglycemia mice model.
