3.5. Data collection

3.5.1. Agronomic Traits

Following agronomic traits were scored on mapping populations. 1. Heading Date HD: number of days required from sowing to the panicles emergence from the leaf sheath. 2. Flag Leaf Length FLL: the length mm from the base to the tip of randomly selected five flag leaves per plant. 3. Flag Leaf width FLW: the width mm of five randomly selected flag leaves measured at the middle of the leaf. 4. Plant height PH: the length cm of stem measured from the soil surface until the tip of highest panicle excluding awn. 5. Panicle Length PL: the length mm from the base of panicle until the tip of the last grain or awn of the grain. Data collected on two panicles were measured from every hill and averaged over two panicles. 6. Panicle weight PW: weight g of two panicles from each plant were weighed. Data collected on two panicles were measured from every hill and averaged over two panicles. 7. Number of total grains per panicle TG: total number of grains of each panicle ere counted. Data collected on two panicles were measured from every hill and averaged over two panicles. 24 8. Seed set SS: percent of filled grains was computed for two panicles. Data collected on two panicles were measured from every hill and averaged over two panicles. 9. Grain weight per plant Wt: weight g of clean filled grains of each plant was recorded. 10. Weight of 100 grains Wt100: weight g of randomly chosen clean and filled 100 grains.

3.5.2. Iron and Zinc Content

The samples were prepared by dehulling using Suzuki-patented Milling Machine made in Brazil. Remaining un-husked grains were removed from the samples. The samples were polished using Kett Grain Polisher Pearlest Machine 110 Volt for three minutes so as to obtain grains with milling degree value of 80 , as measured with whiteness meter. For backcross mapping population, measurement of iron and zinc content was done by ASL Analytical Service Laboratory of IRRI Appendix 8. Two grams of head rice of polished grain samples were sent to ASL for the analysis. Colorimetric the methods Appendix 8 was used to measure iron content of F 2 mapping population.
