DARI BEBERAPA SUMBER AIR PANAS DI
SUMATERA UTARA

ABSTRAK

Telah dilakukan penelitian mengenai isolasi dan karakterisasi parsial dari protease
kasarisolat bakteri termofil dari beberapa sumber air panas di Sumatera
Utara.Sebanyak 20 isolat menunjukkan aktivitas proteolitik secara kualitatif pada
media Skim Milk Agar.Tiga isolat dengan indeks proteolitik terbaik diuji aktivitas
enzimnya. Pengujian aktivitas protease kasar secara kuantitatif menunjukkan bahwa
isolat Bacillus sp. WR02 memiliki aktivitas sebesar 38,01 U/ml pada kondisi pH
6,5 dan suhu 53 oC diikuti oleh isolat Bacillus sp. SQ01 sebesar 13,51 U/ml pada
pH 5 dan suhu 60 oC serta isolat Bacillus sp. KW05 sebesar 13,78 U/ml pada pH 7
dan suhu 53 oC.

PROTEASE ACTIVITY OF THERMOPHILIC BACTERIA ISOLATED FROM
HOT SPRINGS IN SUMATERA UTARA

ABSTRACT

Isolation and partial characterization of crude protease of thermophilic bacteria
from several hot springs in Sumatera Utara has been done. Twenty isolates showed
proteolitic activity qualitatively in Skim Milk Agar medium. Three isolates with
highest proteolitic index were tested their enzyme activity quantitatively. Result
showed that Bacillus sp. WR02 has highest protease activity 38,01 U/ml at pH 6,5
and temperature 53 oC followed byBacillus sp. SQ01 13,51 U/ml at pH 5 and
temperature 60 oC and Bacillus sp.KW05 13,78 U/ml at pH 7 and 53 oC.
