ABSTRAK

Kultur jaringan merupakan teknik perbanyakan tanaman yang sangat
memberi harapan, terutama untuk dunia pemuliaan dalam usaha menghasilkan
bibit unggul dengan kualitas seragam dalam waktu singkat. Salah satu jenis
tanaman yang dapat diperbanyak secara in vitro adalah perbanyakan anggrek
Phalaenopsis sp L. Sampai saat ini tanaman anggrek lebih diminati dibandingkan
tanaman hias lainnya. Pada teknik kultur jaringan pemberian chitosan dapat
meningkatkan pembentukan Protocorn Like Body (PLB) dari eksplan vegetatif
anggrek Phalaenopsis sp L. yang ditumbuhkan pada media padat. Selain itu
penggunaan metode perbanyakan melalui pembentukan PLB ini diharapkan dapat
mengatasi permasalahan produksi tanaman anggrek di Indonesia, yaitu bibit yang
dihasilkan tidak seragam, tidak tergantung waktu dan kualitas bibit sesuai dengan
induknya yang mempunyai sifat superior. Sterilisasi alat dan bahan adalah hal
yang harus diperhatikan dalam penelitian kali ini, sterilisasi alat dapat dilakukan
dengan mengautoclave alat dengan suhu 1210 C selam 2 jam pada tekanan 17,5
psi. Metode penelitian menggunkan Rancangan Acak Lengkap (RAL), dengan
konsentrasi chitosan 0, 5 , 10, 15, 20 dan 25 ppm dengan masing-masing
perlakuan sebanyak 3 ulangan. Dari hasil penelitian dapat diketahui bahwa,
dengan pemberian chitosan 15 ppm pada parameter berat, jumlah dan diameter
PLB menunjukkan hasil yang lebih baik, sedangkan pada konsentrasi 5 ppm pada
parameter jumlah planlet, jumlah daun, jumlah akar dan panjang akar
menunjukkan hasil yang lebih baik dari pada perlakuan lainnya.
