KATA ALUAN KETUA PENGARAH PERKHIDMATAN VETERINAR MALAYSIA
Assalamualaikum WBT, Salam Sejahtera dan Salam 1 Malaysia.
Sektor Penternakan merupakan salah satu sektor penting yang mendapat tumpuan dalam perdagangan dunia kerana ia merupakan pembekal makanan dan sumber protein utama kepada penduduk dunia. Jabatan Perkhidmatan Veterinar (DVS) dengan slogan ‘Tugas Kami Berbakti Kepada Anda’ adalah jabatan yang ditugaskan untuk memastikan bekalan makanan hasil ternakan dalam negara adalah mencukupi dan selamat digunakan. Dengan fungsi utamanya meliputi kesihatan ternakan dan pembangunan komoditi ternakan serta beberapa perkhidmatan sokongan, ini tentunya membuka banyak peluang dan ruang untuk aktiviti penyelidikan dijalankan terutamanya aktiviti penyelidikan gunaan. Selaras dengan itu, penghasilan penyelidikan gunaan adalah digalakkan terutamanya dalam bidang kesihatan dan pengeluaran ternakan. Ini juga merupakan salah satu objektif penting bagi setiap Pusat Ternakan Haiwan di bawah jabatan.
Kerjasama penyelidikan dengan institusi di dalam dan luar jabatan merupakan salah satu strategi DVS untuk meningkatkan kualiti dan kerelevanan penyelidikannya. Sejajar dengan hasrat ini, saya ingin mengucapkan ribuan terima kasih kepada semua yang terlibat terutamanya dari agensi luar kerana bersetuju dan memilih untuk berkerjasama dengan DVS untuk mengembangkan penyelidikan dan inovasi di Negara kita khususnya dalam bidang penternakan. Sesungguhnya ini akan menjadi pemangkin ke arah peningkatan kualiti kesihatan ternakan dan pengeluaran makanan berasaskan ternakan di Malaysia.

Assalamualaikum WBT dan Salam Sejahtera.
Setinggi-tinggi syukur ke hadrat Allah SWT kerana dengan izin-Nya, Kompilasi Senarai Penyelidikan Bahagian Pembangunan Sumber Teknologi Ternakan (BPSTT), Jabatan Perkhidmatan Veterinar (DVS) 2008 – 2014 ini telah berjaya diterbitkan.
Cadangan penerbitan buku kompilasi ini tercetus apabila BPSTT memikirkan bahawa satu bentuk kompilasi penyelidikan adalah perlu untuk memastikan maklumat-maklumat penyelidikan di ladang- ladang DVS dapat direkod dan dikompilasi untuk kemudahan semua pihak.
Adalah diharap dengan terbitnya buku kompilasi ini, ia dapat dijadikan sebagai salah satu sumber rujukan dan penyebaran maklumat informasi-informasi penyelidikan kepada semua pegawai DVS, ahli akademik, pengusaha ternakan dan pihak yang terlibat sama ada secara langsung atau tidak langsung dalam industri penternakan.
Memandangkan penyelidikan merupakan salah satu intervensi yang penting dalam menyokong transfromasi industri ruminan ke arah peningkatan pengeluaran negara, adalah diharap semua pihak yang terlibat dalam bidang penyelidikan untuk mendapat manfaat daripada buku kompilasi ini untuk terus menjalankan penyelidikan menerusi penyelarasan yang lebih berkesan. Penyelarasan penyelidikan adalah perlu bagi memastikan keefiesianan aktiviti penyelidikan dan mengelakkan pertindanan skop-skop penyelidikan.
Saya ingin merakamkan setinggi-tinggi penghargaan kepada Ketua Pengarah DVS atas kebenaran menerbitkan buku ini dan kepada semua kakitangan jabatan dan semua pihak yang telah berusaha untuk menjalankan penyelidikan bersama ini. Tujuan kajian adalah untuk menghasilkan kaedah yang efisien dan murah untuk melakukan Ujian Parentage DNA ke atas lembu dan seterusnya menjadikan Jabatan sebagai sumber rujukan bidang berkaitan. Teknik dan teknologi yang digunakan boleh diaplikasikan dalam membantu pembiakbakaan lembu selain menyelesaikan masalah pertikaian yang mungkin timbul. Ia akan dapat meningkatkan keberkesanan program pembiakbakaan dalam ladang nukleus.
13 penanda genetik mikrosatelit untuk lembu telah diuji.
Penanda genetik mikrosatelit yang sesuai untuk DNA Parentage Testing dalam lembu telah dikenalpasti.
Protokol secara konvensional yang efisien dihasilkan untuk melaksanakan DNA Parentage Testing di Makmal Biologi dan Molekul IBVK.

For the Malaysian Beef Industry to remain competitive and relevant, it is imperative that it uti- lizes the most suitable and efficient of modern beef breeds. One candidate breed that has been identified for strategic sourcing and evaluation is the Piemontese, the most popular among Italian beef breeds. Piemontese cattle are thought to have evolved several millennia ago in the Piemonte region of Italy, having been derived from crosses between the local Aurochs (Bos Taurus) and migrant Zebu cattle from the Indian sub-continent. The Zebu heritage of this breed could render it adaptable to the tropical environment, when used in crossbreeding with local Kedah-Kelantan cattle. Piemontese cattle are known predominantly for their double muscling characteristic, which gives an increase in muscle mass of approximately 20.
The 2 adaptability traits measured were rectal temperature and heart rate. The average rectal temperature of Piedmontese hybrids in IBVK was 38.8 ⁰C (n=4) whereas in PTH Tersat it was 39.0 ⁰C (n=11). This is within the normal body temperature range of 38 - 39.5 ⁰C (Beef Cattle Handbook BCH-3000, Univ. of Arizona, USA 2002). The average heart rate of Piedmontese hybrids in IBVK were 66.0 beats per minute (n=4) and in PTH Tersat it was 73.1 beats per minute. This is within the limits of 48 - 84 beats per minute given in Merck’s Veterinary Manual (2012). There appear to be no adaptability issues with Piedmontese hybrids.
The birth weights of the Piedmontese crosses born in IBVK was 13.0 kg (n=4) and this was similar to that of the purebred Kedah-Kelantan calves born at IBVK. For those born in PTH Tersat the birth weights averaged 13.2 kg (n=11) and this was similar to birth weights of Yellow cattle calves born in Tersat. Birth weights were generally low in these 2 farms as the feeding regime was low input.
This project was initially designed to be conducted at the farmers’ level in the state of Kelantan. However the scientific committee required that the project be conducted in farms under the DVS. The use of artificial insemination technology to produce Piedmontese hybrid cattle for the farmers is being implemented in the state of Kelantan through the State Veterinary Services.

Kedah-Kelantan (KK) cattle are an indigenous cattle breed of Malaysia, which can be found throughout the country but mainly in the northern states of Peninsular Malaysia. The Food And Agriculture Organization, United Nations (FAO) has listed several strategic in situ and ex situ approaches for conservation of indigenous livestock (CGRFAWG AnGR-406Inf. 6, 2006) of which several have been practiced in Malaysia, including: Conservation by continued use in livestock industry production; use of traditional breeds to produce high-value specialty foods and other animal products; maintain traditional landscapes, and support tourism; Cryopreservation of gametes and embryos in national gene banks.
In Malaysia, the process of genetic improvement in livestock in the commercial sector is according to the well-known three-tier genetic improvement pyramid. The breeding policy meeting in 1980 stipulated that pure-line breeding of indigenous Kedah-Kelantan cattle be continued for long-term selection and as a base for future crossbreeding programs. The Department of Veterinary Services (DVS) also, through the National Institute of Veterinary Biodi- versity (NIVB) has been actively cryo-conserving semen from selected top quality genetics of KKs from government and private farms. Although several strategies and actions have been taken by DVS to conserve the KK breed, there are several other measures need to be considered to further strengthen the conservation of the breed in the country as suggested by FAO (CGRFAWG AnGR-406Inf. 6, 2006) :
Development and maintenance of monitoring and warning system for KK breed
Strengthening private-sector capacity to manage the KK breed through establishment of multiplier farms and breed associations.

Traditionally, parentage verification in cattle has been performed using blood typing. Parentage verification is very important, since it is the basis for breeding schemes and becomes even more critical with the widespread use of artificial insemination and embryo transfer techniques. Recently, a new methodology employing DNA markers is being used for parentage control. In order to achieve this goal, information regarding the amount of genetic variation of the microsatellites must be obtained. In the present study, we assessed the polymorphism of 13 microsatellites in regularly reared cattle breed in Malaysia using conventional PCR and genotyping technique. A total of 90 animals belonging to 3 cattle breeds (Kedah-Kelantan, Brahman, Mafriwal) and reared in government Livestock Breeding Center were sampled. Heterozygosity, polymorphism information content, and exclusion probability of the microsatellite were calculated. Considering the 13 microsatellites, exclusion probabilities were as follows: Kedah-Kelantan, 0.5910; Brahman, 0.4506; Mafriwal, 0.5790. All of the thirteen genetic markers in the 3 cattle breeds analyzed, except BM 1824 and ETH 10, were highly informative and their exclusion probabilities demonstrate their usefulness for cattle parentage verification in Malaysia.

Cryopreserved spermatozoa are commonly used in the assisted reproduction procedures such as intrauterine insemination (IUI), in vitro fertilization (IVF) and intraplasmic sperm injection (ICSI). However, spermatozoa plasma membranes functions and integrity are affected by cryopreservation that results in reduced in sperm quality and fertilization rates. Studies have also shown that precentages of apoptotic spermatozoa with impaired functions is much higher after cryopreservation. In ensuring an optimal conception rate rate, marking and elimination of apoptotic spermatozoa are essential. Unfortunately, conventional methods are unable to isolate spermatozoa population based on apoptosis character which is externalization of the membrane phospholipid phosphatidylserine (PS). In this respect, we had used magnetic beads, strong magnetic field and Annexin V – a system known as Magnetic activated cell sorting (MACS) to elim- inate the dead and apoptotic spermatozoa successfully.
Therefore, the objective of this study is to compare the successful rate of conception using the different method of semen preparation and separation. This study is designed to that the promising findings in human spermatozoa can be translated into animal production in Malaysia.

kacukan YC-Limousin. Teknologi AI digunapakai. Baka Limousin mempunyai warna bulu (coat) perang kekuningan dan menghampiri kepa-
da warna ternakan YC. Kemudian kajian kesesuaian pejantan kacukan ini digunakan sebagai pejantan baka untuk peningkatan pengeluaran daging bagi kumpulan YC untuk sejumlah kecil ternakan.

Sebanyak 38 ekor betina YC telah di kenalpasti. Kumpulan Pertama melibatkan 22 ekor betina telah dibiakkan secara AI pada 27.9.12. Didapati 4 ekor anak telah dilahirkan. Kumpulan ke dua sebanyak 16 ekor betina telah dibiakkan secara AI juga pada 26.3.13. Kini sedang menunggu kelahiran sepenuhnya. Kerjasama pegawai dari PTH Tersat, UniSZA dan Unit Pembiakan DVS Ter- engganu amat memuaskan.

DVS telah memperkenalkan pelbagai baka lembu kepada penternak. DVS juga ada menjalankan kajian untuk menilai prestasi baka-baka ini di ladang-ladang Jabatan. Walau bagaimanapun, baka -baka ini tidak dinilai secara sistematik di peringkat penternak. Penilaian baka-baka ini di peringkat penternak adalah sangat penting kerana ini akan memberi petunjuk yang benar berkenaan prestasi baka-baka di ladang-ladang swasta dan sumbangannya kepada produktiviti dan daya saing industri penternakan. Di samping itu, perspektif penternak ke atas sesuatu baka atau baka kacukan dapat memberikan maklumbalas berkenaan potensi sesuatu baka atau baka kacukan dari aspek keuntungan bisnes, kesenangan penternakan, kesesuaian untuk upacara keagamaan (akikahkorban), penerimaan oleh penjual dagingpengguna, dan sebagainya.
Kajian ini dicadangkan supaya prestasi teknikal baka lembu di peringkat penternak dan pembekal dapat ditentukan. Ia juga bertujuan untuk mendokumenkan perspektif penternak berkenaan baka ternakan ruminan yang mereka sendiri telah ternak.
Kaedah yang digunakan dalam kajian ini ialah secara menjalankan survey di lapangan. Sekurang-kurang tiga ratus (300) penternak akan disurvey. Penternak disurvey dipilih secara rambang dari kalangan penternak yang mengusahakan ternakan lembu yang telah berjaya dan telah bertahan dalam industry ini. Input penternak akan direkodkan dengan menggunakan borang survey. Data teknikal setiap ternakan ruminan terdapat di ladang penternak akan direkodkan. Semua maklumat survey dikumpul akan dianalisis untuk menentukan trend dalam persepsi penternak. Data teknikal ternakan dianalisis secara statistikal untuk menentukan prestasinya di la pangan.

Heat stress is a worldwide problem affecting two thirds of the world’s cattle populations. Global warming has resulted in extensive climatic changes in the tropical regions, resulting in increased heat stress to dairy animals of the region. In Malaysia, cows exposed to heat stress from May to September and showed heat shock response because of high temperature. Heat stress occurs in cattle resulting in drastic changes in behavior and physiology including lower growth and low conception rate, reduced feed intake, decreased general activity, increased respiratory and heart beat, panting activity, increased peripheral blood flow and sweating. The severity of heat stress is correlated to ambient temperature, humidity level and solar radiation with little wind. The bovine thermal comfort zone is -13 o
C to +25 o
C. Within this temperature range, the animals comfort is
optimal with body temperature between 38.4 o
C and 39.1 o
C (Lefebvre and Plamondon, 2003) but
the rectal temperature can exceed 41.7 oC in extreme hot weather. In dairy cattle, the optimum temperature is 5 – 20 o
C. Above 25 o
C and even 20 o
C, the animals were suffered from heat stress.
Temperatures in Asian country, especially Malaysia are often much higher than this and gener- ally in the thirties. In dairy cattle, heat stress affects drastic changes in the production of milk and reproductive performance. Fertilization rates in lactating cows decreased under heat stressed
conditions. An increase of about 0.4 °C (0.9 o
F) in a cow’s body temperature has been estimated
to cause 12.8 decline in conception rate in. Heat shock proteins (HSP) are involved in many regulatory pathways and behave as molecular chaperone of cellular proteins and are found in all living organisms from bacteria to humans. The expression of these proteins increased when cells are exposed to elevated temperatures or other stress such as infection, starvation, nitrogen deficiency, exposure cell to toxin and etc. There are a number of HSP families which are categorized according to their molecular weight such as Hsp60, Hsp70, Hsp90, Hsp100 (60, 70, 90, 100 kilodaltons, kDa in size, respectively).

Rumusan Projek Penyakit mastitis merupakan masalah utama kepada pengusaha industri tenusu. Penyakit ini akan menjejas kualiti susu seterusnya mengurangkan pengeluaran serta pendapatan. Pada peringkat ladang, ujian saringan susu dijalankan dengan menggunakan teknik pengiraan somatik sel (somatic cell count, SCC). SCC juga merupakan ujian yang popular dan sering digunakan di ladang pada masa kini untuk mengenalpasti kualiti susu. Walaubagaimanapun, ujian tersebut memberikan tahap sensitiviti yang rendah. Untuk tujuan ini juga, Ujian-ujian lain seperti konduktiviti elektrik (electric conductivity, EC), penanda genetik, serta protein turut dikenalpasti. Namun, teknik-teknik tersebut adalah memakan masa dan memerlukan kos yang tinggi. Penyelidikan ini bercadang untuk mengenalpastikan lembu tenusu yang rentang terhadap penyakit mastitis. Pemilihan akan dijalankan berdasarkan penanda molekul melalui darah dan susu lembu. Dalam menjalankan penyelidikan ini, dua ladang tenusu dikenalpasti bagi mengambil sampel darah dan susu di samping memantau kes mastitis lembu tenusu.

Pada akhir penyelidikan, satu laporan mengenai status jangkitan mastitis ke atas ternakan tenusu di Pusat Ternakan Haiwan, Ayer Hitam akan disediakan kepada pihak DVS. Selain itu, nama-nama pegawai yang terlibat secara langsung dalam penyelidikan ini akan dimasukkan dalam senarai penulis dalam semua penerbitan jurnal yang terhasil daripada usaha kerjasama ini.

Projek dijalankan menggunakan lembu tenusu dari peringkat laktasi awal dan pertengahan. Lem- bu dibahagikan kepada 3 kumpulan, iaitu 1 kumpulan kawalan (tanpa penggunaan lemak bypass dalam makanan) dan lembu yang diberikan makan makanan mengandungi (i) 2.5 lemak bypass dan (ii) 5 lemak bypass dalam makanan kosentrat. Lemak bypass yang digunakan adalah asid stearik yang dihasilkan daripada industri sawit. Makanan diformulasi secara isonitrogenous dan isocaloric. Data pengeluaran susu dicatat setiap hari dan sampel susu dihantar ke Makmal Kawalan Mutu bagi analisis kualiti susu setiap minggu pada hari Rabu. Parameter susu yang diukur ialah kandungan lemak, protein, laktosa, kasein, pepejal tanpa lemak (SNF) dan jumlah pepejal (TS). Da- ta yang diperoleh adalah seperti dalam jadual di bawah. Secara kesimpulan, pemberian lemak by- pass dalam makanan lembu tenusu dapat meningkatkan penghasilan susu.

Proposed study will be held at Pusat Ternakan Haiwan Pantai Timur (PTHPT), 20 cows will be
selected according listed criteria:
1. Apparently healthy
2. Body weight : 220-240 kg
3. Body condition score (1-9 scale) : 4-7
4. Parity : 2-5
5. Breedtype : pure breed of Kedah-Kelantan
6. Reproductive health : regular in cyclicity, proper shape and size of ovary, fallopian tube, uterus
and cervix (determine by rectal palpation)
7. Postpartum status : 80 day past partum and completely involuted uterus Corpus luteum : functional, detectable by rectal palpation
Animal will be divided into two equal groups and each group will be subjected to different proto-
col of estrus synchronization as stated below. Group І
10 cows will be used in estrus synchronization using a controlled internal drug releasing device (CIDR) containing 1.3 mg of progesterone (Pharmacia Limited Company, New Zealand). The de- vice will be inserted to all the cows in group І intravaginally for a period of 8 days. On the 8 th day of CIDR insertion, CIDR will be removed. On day 9, all the cows will be given 1 mg estradiol ben- zoate (Cidoriol, Biomac Lab Ltd) (Malik et.al. 2012). The next day observation of estrus behaviour will be made visually and follicles development through ultrasound machine. This group of cows will be subdivided into two each with 5 cows. One sub group will be paddocked with a teaser bull and the other sub group will be paddock without teaser bull. Group ІІ
Same number of cows as in group І will be treated with PGF2α (25 mg Lutalyse
, Pharmacia
and Upjohn, Kelamazoo, MI) (day 0). On day 2, estrus behaviour of the cows will be observed and follicles development will be observed through ultrasound machine. Similar to group І, 16 cows will
be sub divided into two equal sub groups. One will be paddocked with a teaser bull and another
sub group will be paddock without teaser bull. After monitoring estrus behaviour and ovarian response each of the cows, the cows will be intro-
duced to bull. The next 60 days, the cows will be verified pregnancy through rectal palpation. All of the observation made will be subjected to analysis of variance using SPSS SAS computer soft- ware.

Importance of body size and weight in dairy cattle has been reported to play a major role in milk yield.. Most researchers agree that larger cows have higher milk yield (Erb, 1961 and Clark, 1962). In dairy cattle, phenotypic and genetic relationships between body weight and milk produc- tion have been inconsistent. Therefore this study was conducted to examine the relationship be-
tween metabolic weight and milk production in local Mafriwal dairy cows.
The objective of the study was to identify the relationship between body weight and milk yield
in Mafriwal cows at Ayer Hitam Animal Livestock Centre, Kluang, Johor.
A total of 92 Mafriwal cows were used in the analysis. The milk production of Mafriwal cows was not significantly influenced by cow weight.

The Murrah breed of Water buffalo (Bubalus bubalis) is a breed of domestic water buffalo kept for dairy production. It is originally from Punjab and Haryana states of India and Punjab province of Pakistan. Occasionally jet black in colour, sometimes with white markings on face or legs. Average weight for the males 550 kg and 450 kg females and average milk production is 14 to 15 litres a day.
It was imported into Malaysia recently for the purpose of enhancing the milk production in this country. However, the morphological and performance of this new breed in Malaysia environment has not been thoroughly studied yet. The animals sample selected in this research was from National Institute of Veterinary Biodiversity (NIVB), Jerantut, Pahang and Pusat Ternakan Haiwan (PTH) Air Hitam. A total number of 50 individuals of Murrah buffaloes aged at two years and above will be used in this study.

Somatic cell count (SCC) is an indirect measure of sub-clinical mastitis in dairy cattle. The objective of this study was to evaluate the influence of parity and lactation stage on milk somatic cell counts (SCC) of Mafriwal cows at Air Hitam Livestock Centre, Kluang, and Johor. Milk SCC increased significantly (p<0.05) from 1st lactation (311+54×103 cellsml) to the highest at 5th lactation (936+125×103 cellsml). Similar effect was observed for lactation stage, where milk SCC increased significantly (p<0.05) with days in lactation. The increase of SCC with increased parity and lactation stage might be an indication of the presence of sub-clinical of mastitis.

INTRODUCTION
Cryopreservation is a process where cells, whole tissues or any other substances susceptible to damage caused by chemical reactivity or time are preserved by cooling to sub-zero temperatures. Semen cryopreservation is a procedure to preserve sperm cells. Cryopreservation procedures including cooling, freezing, and thawing induces sperm cell damages derived from the formation of intracellular ice crystals, cold shock and osmotic injury resulting is loss of motility, viability and biochemical changes and finally lead to reduce in fertilizing capability (Maxwell and Salamon, 1995). In general, this study is to identify the effect of plant extracts on chilled and post-thawed quality of cattle bull spermatozoa in extender.
MATHERIALS AND METHOD
Semen Samples will be collected from 2 adult bulls at two week intervals with the aid of an artificial vagina. Immediately after collection, the ejaculates will be stored at 33°C in water bath and transported to the laboratory for semen processing and evaluation. The fresh undiluted ejaculates that containing more than 70 progressively motile spermatozoa will be used.
Semen samples will be diluted at a 1:4 ratio (semen: diluents) to contain 3.5 x 10 spermatozoaml at 37°C. Diluted semen will be divided into five parts and mixed with different parts of plant extract prepared at the Institute of Bioscience, UPM. Diluted semen will be cooled gradually to 5°C within 2 hours, later transferred into 0.25,L straws and will undergo freezing processes using liquid nitrogen.
At every step of semen freezing procedures, semen sample will be analysed for quality (diluted, chilled ans post-thawed). The semen samples will be evaluated using computer assisted sperm analysis (CASA) model IVOS version 12 for motility. Sperm samples also will be stained using eosin-nigrosin (Bearden Fuquay, 1992) to determine the livedead and abnormalities of the s permatozoa.
STATISTICAL ANALYSIS
The percentage of motile, viable, normal or abnormal sperm will be analysed using nonparametric analysis. Differences with values of P < 0.05 are considered to be statistically significant.

Rumusan Projek The results indicated that the mean age at first calving was 1146 ± 148 days, the mean age at conceived was 858 ± 148 days, the mean for first, second, and third calving interval were 403 ± 137, 389 ± 116, and 376 ± 90 days respectively. The result also showed that the calving interval was subsequently decreased with increasing parity. No significant differences were found between three calving intervals (P > 0.05). The mean birth weight was significantly different between fe- male (12.93 kg) and male (13.97 kg) calves (P < 0.05). There was a weak correlation between the calving rate and monthly rainfall pattern (r = 0.26). For mortality rate, there was a weak cor- relation between mortality (below 3 months old) with rainfall pattern (r = 0.17). The results also showed that there is no correlation between mortality below 6 months old (r = -0.28) and total mortality (r = -0.04) with rainfall pattern. In conclusion, Kedah-Kelantan
