The long term goal of this project is aid the conservation of the critically endangered freshwater sawfish
Pristis microdon in Indonesia and throughout its range. Our specific aims are to: (1) determine the
taxonomic status of Indonesian P. microdon; (2) estimate the genetic diversity within populations of P.
microdon in Indonesia and compare this with genetic diversity within Australian populations of the
species; and (3) estimate the genetic differentiation among populations of P. microdon in different areas
of Indonesia and among populations of P. microdon in Indonesia and Australia. To achieve these aims we
will measure the genetic diversity within and among Indonesian populations of P. microdon, using
mtDNA and nuclear microsatellite markers amplified from dried rostra, to avoid the need for sampling
living tissue from these endangered fishes. To date, not much can be reported, as the planned lab works
in the proposal, the activities done were collecting samples (due to the species is belong to the
endangered species the samples are very-very rare, so far we just got at the end of the year 2 samples),
extracting DNA and optimizing PCR for amplification of DNA and also optimizing conditiona for
microsatellite analysis. Two article published, one in an international journal and one article in a
proceeding of national seminar; as well as on in ppt presentation presented in an International seminar.
