YUNITA ARDINI. The Effect of Curcuma xanthorrhiza Roxb. Rhizome Extracts
on the Newborn Rat Cerebrum Cells In Vitro Growth. Under direction of ITA
DJUWITA and MIN RAHMINIWATI.
Research has been conducted on in vitro culture of newborn rat (Sprague
Dawley) cerebrum cells in DMEM (Dulbecco’s Modified Eagle’s Medium)
containing 10% NBCS (Newborn Calf Serum) and 50 µg/mL gentamycin
(mDMEM), with and without Curcuma xanthorrhiza Roxb. rhizome extracts
(CZ). There are five groups of treatment, consisted of positive control
(mDMEM+30 µg/mL asiaticoside (AC)), negative control (mDMEM), CZ 100
ppm (mDMEM+100 ppm CZ), CZ 200 ppm (mDMEM+200 ppm CZ), and CZ
400 ppm (mDMEM+400 ppm CZ). Culture was done in 5% CO2 incubator at
37°C for 6 days. The parameters observed were Population Doubling Time (PDT),
neuron and glia composition, and the length of axon and dendrite, were done
based on calculation using hemocytometer, Hematoxylin Eosin (HE) staining, and
measured using micrometer, respectively. Data were analyzed using statistical
ANOVA and Duncan. The results showed that Curcuma xanthorrhiza Roxb.
rhizome extracts concentration 100 ppm inhibited the neuronal cells proliferation
