AKTIVITAS ANTIBAKTERI FRAKSI DARI EKSTRAK ETANOL UMBI BATANG TUMBUHAN SARANG SEMUT (Myrmecodia pendens Merr & L.M. Perry) TERHADAP Klebsiella pneumoniae DAN Shigella dysenteriae.
ABSTRAK
Klebsiella pneumoniae merupakan bakteri penyebab pneumonia yang dapat
mengawali terjadinya infeksi lain seperti disentri yang disebabkan oleh infeksi
bakteri Shigella dysenteriae. Tumbuhan sarang semut (Myrmecodia pendens
Merr. & L.M. Perry) secara empiris banyak digunakan untuk mengobati berbagai
macam penyakit. Berdasarkan penelitian sebelumnya ekstrak etanol sarang semut
diketahui memiliki aktivitas antibakteri terhadap Klebsiella pneumoniae dan
Shigella dysenteriae. Pada penelitian ini dilakukan uji aktivitas antibakteri dari
fraksinya. Ekstraksi dilakukan dengan metode maserasi menggunakan etanol 70%.
Fraksinasi dilakukan dengan Kromatografi Cair Vakum menggunakan eluen
gabungan n-heksana, etil asetat, dan metanol dengan gradien kepolaran
meningkat. Penggabungan fraksi dipantau dengan Kromatografi Lapis Tipis
berdasarkan pola bercak yang sama. Pengujian aktivitas antibakteri fraksi
dilakukan dengan metode microdilution dan hasilnya menunjukan bahwa seluruh
fraksi memiliki aktivitas terhadap kedua bakteri uji. Fraksi teraktif untuk
Klebsiella pneumoniae terletak pada fraksi E, yaitu fraksi n-heksana dan etil asetat
(10:90) dengan nilai Konsentrasi Bunuh Minimum (KBM) 0,125% (b/v). Fraksi
teraktif untuk Shigella dysenteriae terletak pada fraksi F, yaitu fraksi etil asetat
dan metanol (90:10) dengan nilai KBM 0,055% (b/v).

ABSTRACT
Klebsiella pneumoniae is a bacteria causing pneumonia and the occurrence
initiator of other infection, such as dysentery caused by Shigella dysenteriae
bacterial infection. Ant plant (Myrmecodia pendens Merr. & L.M. Perry) are
empirically used to cure many kinds of disease. Based on the earlier research,
ethanol extract of ant plant has antibacterial activity against Klebsiella
pneumoniae and Shigella dysenteriae. This research was conducted to determine
the antibacterial activity of its fractions. Extraction was conducted using
maceration method with 70% ethanol as the solvent. Fractionation was conducted
using Liquid Vacuum Chromatography with combination of n-hexane, ethyl
acetate and methanol that had the increasing polarity gradient as the eluent.
Fractions merging were monitored using Thin layer Chromathography based on
the same spot pattern. Antibacterial activity test was done using agar
microdilution method and the result of the activity test showed that all fractions
have activity against both bacteria. The most active fraction against Klebsiella
pneumoniae is fraction E that containing n-hexane and ethyl acetate fraction
(10:90) with Minimum Bactericidal Concentration (MBC) value 0,125% (w/v).
The most active fraction against Shigella dysenteriae is fraction F that containing
ethyl acetate and methanol fraction (90:10) with MBC value 0,055% (w/v).
