within Peranakan Etawah, Saanen and PESA Goat

This research was conducted to identify genetic polymorphism at the exon 4 growth
hormone gene in three goat breeds, namely PE, Saanen and PESA. Total DNA
samples used are 240 samples which consist of three breeds, namely PE (98 samples,
from Ciapus 20 samples, Carius 28 samples and Sukajaya 50 samples), Saanen (91
samples, from Cijeruk 20 samples, Cariu 31 samples and Sukabumi 40 samples) and
PESA (51 samples, from Cijeruk 7 samples, Cariu 25 samples and Balitnak 19
samples). The GH gene exon 4 amplified by thermocylcer machine at 64 oC with 200
base pair length. The PCR product was analyzed by SSCP method using 10% of
acrylamide concentration for 8 hours at 250 voltage. The result showed there are four
conformational patterns with each genotype frequency, they are DD (0,211), DE
(1,000), EE (0,600) and GH (1,000). The SSCP analysis result found four alleles
with their highest frequencies, namely D (0,605) in PESA breed, E (0,800) in PE
breed, G and H (0,500) in Saanen and PESA breed. The result of χ² test showed that
the growth hormone gene exon 4 are in Hardy-Weinberg disequilibrium generally in
