Streptococcus mutans merupakan bakteri yang berperan penting dalam proses
pembentukan karies gigi. Bakteri ini merupakan flora normal rongga mulut yang
dapat menjadi patogen bila terjadi peningkatan jumlah koloni yang berlebihan. Kopi
merupakan salah satu tanaman hasil perkebunan di kota Jember. Selain dikonsumsi
sebagai minuman, kopi juga dimanfaatkan dalam bidang kesehatan, karena
kandungan dalam biji kopi yang memiliki daya hambat. Tujuan dari penelitian ini
adalah untuk mengetahui daya hambat ekstrak biji kopi Robusta terhadap
pertumbuhan S. mutans.
Jenis penelitian ini adalah eksperimental laboratoris dengan rancangan
penelitian the post test only control group design. Sampel terbagi dalam tujuh
kelompok perlakuan yaitu kelompok R100, R50, R25, R12,5, Bet K(+), Kaf K(+) dan
K(-). Bahan-bahan perlakuan pada kelompok tersebut dimasukkan ke dalam lubang
sumuran 12 petridish yang berisi media BHI-A yang telah terinokulasi S. mutans
sesuai dengan kode kelompoknya. Seluruh petridish dimasukkan kedalam desikator
dan diinkubasi dalam inkubator pada suhu 37°C selama 24 jam. Setelah 24 jam
dilakukan pengamatan dan pengukuran dengan menggunakan jangka sorong.
Data hasil penelitian dianalisis. Secara deskripsi menunjukkan bahwa
kelompok R100 mempunyai rata-rata diameter zona hambat paling besar yaitu
sebesar 10,3 mm dan rata-rata diameter zona hambat terkecil pada kelompok Kaf
(K+) sebesar 7 mm. Uji statistik Kruskal Wallis menunjukkan ada perbedaan yang
bermakna pada semua kelompok. Untuk mengetahui kelompok yang berbeda
bermakna dilanjutkan dengan uji Mann-Whitney. Hasil uji menunjukkan bahwa

terdapat perbedaan yang bermakna pada semua kelompok, kecuali pada kelompok
R50 dengan kelompok R25, kelompok R25 dengan kelompok R12,5, kelompok R25
dengan kelompok Bet K(+), kelompok R12,5 dengan kelompok Bet K(+), kelompok
R12,5 dengan kelompok Kaf K(+) dan kelompok Bet K(+) dengan Kaf K(+)
memiliki perbedaan yang tidak bermakna.
Hasil dari penelitian ini dapat disimpulkan bahwa ekstrak biji kopi Robusta
(Coffea robusta) mempunyai daya hambat terhadap S. mutans. Konsentrasi terkecil
dari ekstrak biji kopi Robusta (Coffea robusta) yang masih mempunyai daya hambat
adalah konsentrasi 12,5%.
