Abstract
Ba ckg rou nd : Seed ext ract of Swiet enia m ahagoni ( L.) Jacq. cont ains saponins,
flavonoids, alkaloids, t erpenoids, st eroids, and t annins t hat m ay exhibit significant
prot ect ion of kidney cells from free radicals. I n present st udy, Swiet enia m ahagoni
was evaluat ed for it s nephroprot ect or effect on paracet am ol- induced kidney cells
hist ological dam age in m ice ( Mus m usculus) .

M e t ho ds: This resear ch used experim ent al laborat ory st udies w it h post t est only
cont rolled group design. Sam ples were 28 m ale Swiss webst er m ice ( 2- 3 m ont hs
old) weighing + 20 g and divided int o 4 groups, 7 m ice each group. Sam pling
t echnique in t his resear ch w as incident al sam pling. The negat ive cont rol group ( KK
( - ) ) and t he posit ive cont rol group ( KK ( + ) ) of m ice were adm inist ered aquadest for
14 days. The first t reat m ent group ( KP1 ) and t he second group t reat m ent ( KP2 ) of
m ice were adm inist ered m ahagony seed ext ract w it h t he dose of 11,2 m g/ 20 g and
22,4 m g/ 20 g body w eight of m ice for 14 days, respect ively. Paracet am ol was
adm inist er ed t o groups of KK ( + ) , KP1 , dan KP2 on t he 12 t h , 13 t h , and 14 t h day. On
th

day- 15 , m ice were sacrificed and t heir kidneys were t aken and st ained wit h
hem at oxilin eosin ( HE) for hist ological st udy. The kidney cells hist ological feat ures
were assessed based on quant ifying of pyknosis, karyorrhexis, and karyolysis. Dat a
were analyzed wit h t he One- Way ANOVA test (α = 0.05) and Post Hoc Mult iple
Com parisons LSD test (α = 0.05).
Re sult s: The m ean of kidney cells hist ological dam age in m ice for KK ( - ) , KK ( + ) ,
KP1 and KP2 were 9.57 + 0.701; 28.93 + 1.698; 9.28 + 0.873 and 27.79 + 1.651,
respect ively. Result of st at ist ic analysis showed t hat t here w er e significant
differences of kidney cells dam age score bet ween KK ( - ) – KK ( + ) , KK ( - ) – KP2 , KK

( + ) – KP1 , KP1 – KP2 , and non significant differences bet ween KK ( - ) – KP1 and KK
( + ) – KP2 .
Co nclusio ns: Swiet enia m ahagoni ( L.) Jacq. seed ext ract showed nephr oprot ect or
effect against paracet am ol- induced kidney cells hist ological dam age in m ice.
