1. Pembuatan Seduhan Teh Hijau dan Teh Hitam
Ditimbang teh hijau dan teh
hitam sebanyak 1750 mg/kg
bb, 3500 mg/kg bb dan 7000
mg/kg bb
Diseduh dengan air mendidih sebanyak 30 ml selama 3 menit
lalu disaring menggunakan kain flanel
Seduhan teh dosis1750
mg/kg bb, 3500 mg/kg
bb dan 7000 mg/kg bb
2. Pengujian metode defekasi
36 ekor mencit
Diberi penandaan pada masing-masingekor mencit
Ditimbang dan catat berat badanmencit
Diberikan perlakuansecara per oral: suspensi CMC 1%, teh
Hijau, teh Hitam dan suspensiloperamid HCl 2 mg
30 menit kemudian diberikan Oleum ricini 0,5 ml secara p.o
Diamati selama enam jam dengan interval waktu tiap 30 menit.

36 ekor mencit
Ditimbang dan catat berat badanmencit
Diberikan perlakuansecara per oral : suspensi CMC 1%, teh
Hijau, teh Hitam dan suspensiloperamid HCl 2 mg
30 menit kemudian diberikan Oleum ricini 0,5 mlsecara p.o
dibiarkan kembali selama 30 menit, Kemudian semua mencit
dikorbankan dengan cara dislokasi leher
dilakukan pembedahan. Saluran gastroinstetinal dari phylorus
sampai caecum dipotong
isinya dikeluarkan dan ditampung dalam gelas ukur untuk
ditimbang volumenya

Perhitungan dosis CMC Na 1%
=
= 10 mg/ml
Perhitungan CMC Na 1% pada mencit 25 g
=
x 25 g = 0,25 ml
Volume yang diberikan = 0,25 ml
b) Perhitungan dosis Lodia® tablet
Tiap tablet Lodia mengandung 2 mg Loperamid HCl
Dosis dewasa 2 mg – 8 mg/hari dan tidak lebih dari 16 mg/hari (berat dewasa
untuk konversi 70 Kg).
Dosis untuk mencit 20 g yaitu :
(2 mg– 8 mg) x 0,0026 = 0,0052 – 0,0208 mg/hari dan tidak lebih dari 0,04616
mg/Kg bb.
Berat serbuk Lodia yang akan diambil yaitu dari 20 tablet lodia® di gerus
dan ditimbang = 2136 mg.
Dari 20 tablet mengandung loperamid HCl sebanyak 40 mg.
Dosis loperamid HCl yang digunakan adalah dosis maksimum yaitu :
= 0,0416 mg/20 g bb mencit
= 2,08 mg/kgbb
Loperamid HCl yang digunakan adalah : 2,08 mg/kg bb

Maka serbuk loperamid yang di gunakan adalah :

Dalam serbuk lodia® sebanyak 111,07
mg
mengandung
loperamid HCl
sebanyak 2,08 mg.
Suspensi loperamid HCl yang
akan dibuat adalah 111,07 serbuk lodia®
dilarutkan dalam 10 ml suspensi CMC 1 %.
c) Perhitungan dosis seduhan teh hijau dan teh hitam
1. Untuk dosis 1750 mg/kg bb
Jumlah yang diberikan =
x 25 g = 43,75 mg
Jumlah seduhan yang diberikan =
x 30 ml = 0,75 ml
2. Untuk dosis 3500 mg/kg bb
Jumlah yang diberikan =
x 25 g = 87,5 mg
Jumlah seduhan yang diberikan =
x 30 ml = 0,75 ml
3. Untuk dosis 7000 mg/kg bb
Jumlah yang diberikan =

1. Pembuatan Seduhan Teh Hijau dan Teh Hitam
Ditimbang teh hijau dan teh
hitam sebanyak 1750 mg/kg
bb, 3500 mg/kg bb dan 7000
mg/kg bb
Diseduh dengan air mendidih sebanyak 30 ml selama 3 menit
lalu disaring menggunakan kain flanel
Seduhan teh dosis1750
mg/kg bb, 3500 mg/kg
bb dan 7000 mg/kg bb
2. Pengujian metode defekasi
36 ekor mencit
Diberi penandaan pada masing-masingekor mencit
Ditimbang dan catat berat badanmencit
Diberikan perlakuansecara per oral: suspensi CMC 1%, teh
Hijau, teh Hitam dan suspensiloperamid HCl 2 mg
30 menit kemudian diberikan Oleum ricini 0,5 ml secara p.o
Diamati selama enam jam dengan interval waktu tiap 30 menit.

Ditimbang dan catat berat badanmencit
Diberikan perlakuansecara per oral : suspensi CMC 1%, teh
Hijau, teh Hitam dan suspensiloperamid HCl 2 mg
30 menit kemudian diberikan Oleum ricini 0,5 mlsecara p.o
dibiarkan kembali selama 30 menit, Kemudian semua mencit
dikorbankan dengan cara dislokasi leher
dilakukan pembedahan. Saluran gastroinstetinal dari phylorus
sampai caecum dipotong
isinya dikeluarkan dan ditampung dalam gelas ukur untuk
ditimbang volumenya

b) Perhitungan dosis Lodia® tablet
Tiap tablet Lodia mengandung 2 mg Loperamid HCl
Dosis dewasa 2 mg – 8 mg/hari dan tidak lebih dari 16 mg/hari (berat dewasa
untuk konversi 70 Kg).
Dosis untuk mencit 20 g yaitu :
(2 mg– 8 mg) x 0,0026 = 0,0052 – 0,0208 mg/hari dan tidak lebih dari 0,04616
mg/Kg bb.
Berat serbuk Lodia yang akan diambil yaitu dari 20 tablet lodia® di gerus
dan ditimbang = 2136 mg.
Dari 20 tablet mengandung loperamid HCl sebanyak 40 mg.
Dosis loperamid HCl yang digunakan adalah dosis maksimum yaitu :
= 0,0416 mg/20 g bb mencit
= 2,08 mg/kgbb
Loperamid HCl yang digunakan adalah : 2,08 mg/kg bb
