ABSTRAK
Salah satu cara meningkatkan kesehatan tubuh adalah dengan menjaga
kelancaran pengeluaran urin. Pengeluaran urin yang tidak lancar pada kandung
kemih atau ginjal dalam waktu yang lama akan menimbulkan pengkristalan dari
zat-zat yang akan dibuang. Secara medis sudah banyak obat kimia yang
digunakan sebagai diuretika, selain obat kimia banyak tumbuhan yang digunakan
sebagai diuretik, salah satunya tumbuhan senduduk (Melastoma malabathricum
Linn.).
Tujuan penelitian ini adalah untuk mengetahui efek diuretik ekstrak etanol
daun senduduk (EEDS) dengan parameter volume urin, kadar natrium dan kadar
kalium dalam urin tikus jantan.
Hewan yang digunakan adalah tikus jantan sebanyak 25 ekor. Sebelum
dilakukan percobaan tikus terlebih dahulu dipuasakan selama ± 18 jam kemudian
diberikan NaCl 0,9% secara oral sebanyak 20 ml/kg bb sebagai loadingdose.
Tikus dibagi menjadi 5 kelompok yaitu kontrol negatif diberi CMC-Na 0,5%,
pembanding digunakan furosemid dosis 10 mg/kg bb, dan sediaan uji dengan
variabel dosis 400, 500, 600 mg/kg bb. Tikus diletakkan dalam kandang
metabolik. Volume urin yang diekskresikan dicatat setiap jam dan akumulasinya
selama 5 jam sebagai urin total, selanjutnya diukur kadar natrium dan kalium
dengan menggunakan Atomic Absorption Spectrophotometer.
Hasil penelitian menunjukkan bahwa pemberian EEDS dosis 400, 500,
600 mg/kg bb meningkatkan volume urin, kadar natrium dan kalium. Diperoleh
rata-rata volume urin total kontrol negatif, EEDS dosis 400, 500, 600 mg/kg bb,
dan pembanding masing-masing 1,34 ± 0,18 ml; 2,2 ± 0,30 ml; 2,7 ± 0,25 ml;
3,18 ± 0,21 ml; 3,88 ± 0,22 ml; dengan nilai signifikansi (p < 0,05) terhadap
kontrol negatif dan pembanding. Kadar natrium rata-rata masing-masing 31,97 ±
3,78 meq/l; 50,32 ± 3,06 meq/l; 58,30 ± 5,24 meq/l; 77,74 ± 3,91 meq/l; 96,81 ±
3,96 meq/l; dengan nilai signifikansi (p < 0,05) terhadap kontrol negatif dan
pembanding. Kadar kalium rata-rata masing-masing 19,42 ± 2,53 meq/l; 27, 82 ±
1,76 meq/l; 33,83 ± 4,70 meq/l; 39,25 ± 2,25 meq/l; 51,29 ± 4,11 meq/l ; dengan
nilai signifikansi (p < 0,05) terhadap kelompok kontrol negatif dan pembanding.
Berdasarkan hasil di atas dapat disimpulkan bahwa EEDS dosis 400, 500,
600 mg/kg memberikan efek diuretik dalam meningkatkan volume urin, kadar
natrium dan kadar kalium dalam urin tikus jantan dengan efek yang paling baik
ditunjukkan oleh dosis 600 mg/kg bb.

ON MALE RATS
ABSTRACT
One way to improve the health of the body is to maintain the smooth
passage of urine, urine output is not smooth in the bladder or kidneys in a long
time will cause crystallization of substance to be scrapped. In the medical world
has many chemical drugs produced as a diuretic, other than chemical drugs many
medicinal plants that are used as a diuretic, one of which is senduduk
(Melastomamalabathricum Linn.).
The purpose of this study was to determine the diuretic effect of ethanol
extract of leaves senduduk parameters urine volume, sodium and potassium levels
in the urine of male rats.
Animals used were male rats. Rats were fasted for ± 18 hours and then
given 0.9% NaCl administrated orally at a dose of 20 ml/kg bw. Rats were divided
into 5 groups: negative control group given 0.5% CMC-Na, the positive control
was given furosemide 10 mg/kg bw, and the administration of the ethanol extract
of the leaves ingredients senduduk doses of 400, 500, 600 mg/kg bw. Rats placed
in metabolic cages that have been modified. Excreted urine volume and
accumulation recorded every hour for 5 hours as the total urine, and determined
the levels of electrolytes sodium and potassium in the urine using by Atomic
Absorption Spectrophotometer.
The results showed that administration of a dose EEDS 400, 500, 600
mg/kg bw increase urine volume, sodium and potassium. Gained an average total
urine volume negative control, EEDS doses of 400, 500, 600 mg/kg bw, and
comparison respectively 1,34 ± 0,18 ml; 2.2 ± 0.30 ml; 2.7 ± 0.25 ml; 3.18 ± 0.21
ml; 3.88 ± 0.22 ml, with a value of significance (p < 0.05) to the negative control
and comparison. Average sodium levels respectively 31.97 ± 3.78 meq/l; 50.32 ±
3.06 meq/l; 58.30 ± 5.24 meq/l; 77.74 ± 3.91 meq/l; 96.81 ± 3.96 meq/l, with a
value of significance (p < 0.05) to the negative control and comparison. Average
potassium levels respectively 19.42 ± 2.53 meq/l; 27.82 ± 1.76 meq/l; 33.83 ±
4.70 meq/l; 39.25 ± 2.25 meq/l; 51.29 ± 4.11 meq/l, with a value of significance
(p < 0.05) to the negative control and comparison.
Based on the above results EEDS doses of 400, 500, 600 mg/kg bw gives a
diuretic effect in increasing urine volume, sodium and potassium levels in the
urine of male rats to the most well demonstrated by a dose of 600 mg/kg bw.
