Analisis SSR (Simple Sequence Repeats) Pada Kelapa Sawit (Elaeis guineensis Jacq.) Asal Klon Plasma Nutfah PT.Socfindo

ABSTRACT

DIAN ARVITA: SSR Analysis’ (Simple Sequence Repeats) of Oil Palm from
Germplasm Clone of PT.Socfindo. Supervised by LOLLIE AGUSTINA P.PUTRI
and EVA SARTINIBAYU.
The purpose of this research was to find out band pattern of Oil Palm Clone
based on Simple Sequence Repeats with marker FR 0783, FR 0779, FR 3663, and
FR 3745. This research was conduted in genetica laboratory of molekuler,
Faculty of Agriculture, University of Sumatera Utara and DNA laboratory
Central Selection Bangun Bandar of PT.Socfindo, Dolok Masihol Regency, North
Sumatera from March 2016 to June 2016. The materials in this research come
form Germplasm Clone of PT.Socfindo. The number of materials in this research
was 30. DARwin 6.0 and MicrosoftExcel 2007 was used calculated and describe
analysis. The result of this research showed that FR 0779 is a monomorphic
primer at 320 bp and 243 bp, FR 3663 is a monomrphic primer at 242 bp. While,
FR 0783 is a polymorphiv primer at 335 bp, 272 bp, 351bpand 300 bp, FR 3745
is a polymorphic primer at 317 bp and 264 bp. FR 3745 showed difference band
pattern of DNA at sampel number 18 and FR 0783 at sampel number 29

DIAN ARVITA : Analisis SSR (Simple Sequence Repeats) pada Kelapa Sawit

Penelitian ini bertujuan untuk mengetahui pola pita DNA klon kelapa sawit
berdasarkan marka Simple Sequence Repeats dengan primer FR 0783, FR 0779,
FR 3663, dan FR 3745. Penelitian ini telah dilakukan di laboratorium genetika
molekuler Fakultas Pertanian Universitas Sumatera Utara dan laboratorium DNA
Pusat Seleksi Bangun Bandar PT. Socfindo pada Maret - Juni 2016. Bahan
tanaman yang digunakan berasal dari klon plasma nutfah PT.Socfindo sebanyak
30 klon. Perhitungan dan analisis deskriptif pada penelitian ini menggunakan
sotware DARwin 6.0 dan Microsoft Excel 2007. Hasil penelitian menunjukkan
bahwa Primer monomorfis ditunjukkan oleh FR 0779 pada 320 bp dan 243 bp
serta FR 3663 pada 242 bp sedangkan primer polimorfis ditunjukkan oleh FR
0783 pada 335 bp, 272 bp, 351 bp dan 300 bp serta FR 3745 pada 317 bp dan 264
bp. Primer FR 3745 dapat menunjukkan perbedaan pola pita DNA pada sampel
sampel No. 18 serta primer FR 0783 yaitu pada sampel No. 29.
