RINGKASAN
Ekstrak Etanol Daun Benalu (Macrosolen cochinchinensis) Sebagai Peningkat
Jumlah Trombosit pada Tikus Putih (Rattus norvegicus) yang Terinfeksi Virus
Dengue ; Rr. Okiningtyas Kun Cahyandari, 072010101014; 2010; 40 hlm; Fakultas
Kedokteran Universitas Jember.
Demam Berdarah Dengue (DBD) adalah suatu penyakit infeksi akibat virus
dengue yang ditularkan melalui gigitan nyamuk Aedes aegypti (A. aegypti). Penyakit
ini dapat bermanifestasi sebagai Dengue Shock Syndrome (DSS) yang bisa
mengakibatkan syok dan berujung pada kematian. Salah satu penyebab kematian
pada DBD adalah penurunan jumlah trombosit (trombositopeni) yang terjadi akibat
meningkatnya agregasi trombosit akibat endotel vaskuler yang rusak, destruksi
trombosit dan menurunnya proses trombopoesis.
Meskipun DBD sudah menjadi penyakit serius di Indonesia selama puluhan
tahun, sampai saat ini masih belum ditemukan obat maupun vaksin yang dapat
menyembuhkan penyakit DBD tersebut. Pengobatan yang dilakukan saat ini hanya
bersifat simptomatis dan bertujuan untuk mengganti plasma yang hilang disebabkan
kebocoran plasma dengan cara memberi terapi cairan. Akan tetapi, terapi cairan
sendiri mempunyai beberapa efek samping seperti edema dan asidosis laktat. Oleh
karena itu, perlu ditemukan obat-obatan spesifik yang bersifat kausatif yang bisa
menyembuhkan penyakit DBD baik dari segi farmakologis maupun bahan alami.
Penelitian ini menggunakan daun benalu (Macrosolen cochinchinensis) yang
telah diekstraksi menjadi ekstrak etanol dan diduga memiliki efek trombopoeisis.
Selain itu, senyawa quersetin yang terkandung dalam daun benalu berfungsi

penelitian ini adalah membuktikan secara ilmiah khasiat ekstrak etanol daun benalu
(Macrosolen cochinchinensis) sebagai peningkat jumlah trombosit pada tikus putih
(Rattus norvegicus) yang terinfeksi virus dengue.
Sampel terdiri dari 24 ekor tikus Rattus norvegicus jantan yang dipilih secara
acak dan diinfeksi dengan virus dengue serotip DEN-2 serta dibagi ke dalam empat
kelompok yaitu tiga kelompok perlakuan dengan pemberian ekstrak etanol daun
benalu berbagai dosis yang masing-masing 37,5 mg/100grBB; 75 mg/100grBB dan
150 mg/100grBB serta satu kelompok kontrol dengan pemberian aquadest. Variabel
yang diukur adalah jumlah trombosit dalam darah tikus. Pengukuran jumlah
trombosit dalam darah tikus menggunakan metode blood analyzer.
Uji analisa data menggunakan One Way Anova pada α = 0,05, menunjukkan
hasil bahwa terdapat perbedaan yang bermakna antara ketiga kelompok perlakuan
dimana nilai signifikansi lebih kecil dari 0,05. Kandungan bahan dalam ekstrak etanol
daun benalu yang dapat meningkatkan jumlah trombosit adalah senyawa quersetin
yang berperan sebagai trombopoesis dan terbukti menurunkan permeabilitas vaskular
sehingga mampu mencegah terjadi kebocoran plasma. Penurunan permeabilitas
kapiler ini menyebabkan trombosit tidak terpakai untuk menutupi endotel vaskuler
yang rusak sehingga jumlah trombosit akan kembali meningkat. Dengan demikian,
ekstrak etanol daun benalu (Macrosolen cochinchinensis) dapat dikembangkan lebih

RINGKASAN
Ekstrak Etanol Daun Benalu (Macrosolen cochinchinensis) Sebagai Peningkat
Jumlah Trombosit pada Tikus Putih (Rattus norvegicus) yang Terinfeksi Virus
Dengue ; Rr. Okiningtyas Kun Cahyandari, 072010101014; 2010; 40 hlm; Fakultas
Kedokteran Universitas Jember.
Demam Berdarah Dengue (DBD) adalah suatu penyakit infeksi akibat virus
dengue yang ditularkan melalui gigitan nyamuk Aedes aegypti (A. aegypti). Penyakit
ini dapat bermanifestasi sebagai Dengue Shock Syndrome (DSS) yang bisa
mengakibatkan syok dan berujung pada kematian. Salah satu penyebab kematian
pada DBD adalah penurunan jumlah trombosit (trombositopeni) yang terjadi akibat
meningkatnya agregasi trombosit akibat endotel vaskuler yang rusak, destruksi
trombosit dan menurunnya proses trombopoesis.
Meskipun DBD sudah menjadi penyakit serius di Indonesia selama puluhan
tahun, sampai saat ini masih belum ditemukan obat maupun vaksin yang dapat
menyembuhkan penyakit DBD tersebut. Pengobatan yang dilakukan saat ini hanya
bersifat simptomatis dan bertujuan untuk mengganti plasma yang hilang disebabkan
kebocoran plasma dengan cara memberi terapi cairan. Akan tetapi, terapi cairan
sendiri mempunyai beberapa efek samping seperti edema dan asidosis laktat. Oleh
karena itu, perlu ditemukan obat-obatan spesifik yang bersifat kausatif yang bisa
menyembuhkan penyakit DBD baik dari segi farmakologis maupun bahan alami.
Penelitian ini menggunakan daun benalu (Macrosolen cochinchinensis) yang
telah diekstraksi menjadi ekstrak etanol dan diduga memiliki efek trombopoeisis.
Selain itu, senyawa quersetin yang terkandung dalam daun benalu berfungsi

penelitian ini adalah membuktikan secara ilmiah khasiat ekstrak etanol daun benalu
(Macrosolen cochinchinensis) sebagai peningkat jumlah trombosit pada tikus putih
(Rattus norvegicus) yang terinfeksi virus dengue.
Sampel terdiri dari 24 ekor tikus Rattus norvegicus jantan yang dipilih secara
acak dan diinfeksi dengan virus dengue serotip DEN-2 serta dibagi ke dalam empat
kelompok yaitu tiga kelompok perlakuan dengan pemberian ekstrak etanol daun
benalu berbagai dosis yang masing-masing 37,5 mg/100grBB; 75 mg/100grBB dan
150 mg/100grBB serta satu kelompok kontrol dengan pemberian aquadest. Variabel
yang diukur adalah jumlah trombosit dalam darah tikus. Pengukuran jumlah
trombosit dalam darah tikus menggunakan metode blood analyzer.
Uji analisa data menggunakan One Way Anova pada α = 0,05, menunjukkan
hasil bahwa terdapat perbedaan yang bermakna antara ketiga kelompok perlakuan
dimana nilai signifikansi lebih kecil dari 0,05. Kandungan bahan dalam ekstrak etanol
daun benalu yang dapat meningkatkan jumlah trombosit adalah senyawa quersetin
yang berperan sebagai trombopoesis dan terbukti menurunkan permeabilitas vaskular
sehingga mampu mencegah terjadi kebocoran plasma. Penurunan permeabilitas
kapiler ini menyebabkan trombosit tidak terpakai untuk menutupi endotel vaskuler
yang rusak sehingga jumlah trombosit akan kembali meningkat. Dengan demikian,
ekstrak etanol daun benalu (Macrosolen cochinchinensis) dapat dikembangkan lebih
