UJI TOKSISITAS AKUT EKSTRAK ETANOL DAUN LIDAH MERTUA
(Sansevieria trifasciata Prain) TERHADAP MENCIT JANTAN
ABSTRAK
Daun Lidah Mertua (Sansevieria trifasciata Prain) umumnya dikenal dan
dibudidayakan oleh masyarakat sebagai tanaman hias. Selain itu, tanaman ini juga
dapat digunakan sebagai antidiabetes, antibakteri, analgesik, antipiretik, dan
antioksidan. Tujuan dari penelitian ini adalah untuk mengetahui potensi
ketoksikan akut ekstrak etanol daun lidah mertua.
Penelitian ini merupakan penelitian eksperimen dengan menggunakan 30
ekor mencit jantan yang dibagi menjadi 1 kelompok kontrol dan 5 kelompok
perlakuan, masing-masing terdiri dari 5 ekor mencit. Kelompok kontrol diberi
suspensi CMC-Na 0,5%, sedangkan kelompok perlakuan diberi suspensi sediaan
uji ekstrak etanol daun lidah mertua dengan dosis 250, 500, 1000, 2000, dan 4000
mg/kg bb. Sediaan uji diberikan secara oral dengan hanya satu kali pemberian
pada awal masa penelitian. Pengamatan intensif dilakukan selama 4 jam pertama,
meliputi gejala toksik yang muncul dan jumlah mencit yang mati selama 14 hari,
serta kajian histopatologi pada organ hati dan ginjal. Data yang diperoleh
kemudian dianalisis dengan menggunakan ANOVA satu arah dan dilanjutkan
dengan uji Post Hoc, yaitu LSD dan Tukey.
Hasil uji toksisitas menunjukkan bahwa mencit yang diberikan ekstrak
etanol daun lidah mertua pada kelompok kontrol dan kelompok perlakuan dengan

dosis 250, 500, dan 1000 mg/kg bb tidak menunjukkan gejala toksik, namun pada
kelompok perlakuan dengan dosis 2000 dan 4000 mg/kg bb menunjukkan gejala
toksik pada uji panggung serta adanya aktivitas berjalan dengan menyeret perut
tetapi tidak ada mencit yang mati. Dari hasil pengamatan, ekstrak etanol daun
lidah mertua pada dosis 4000 mg/kg bb memberikan pengaruh terhadap kadar
SGPT (p < 0,05). Selain itu, terjadi kerusakan / nekrosis sel hati pada kelompok
perlakuan dengan dosis 500, 1000, 2000, dan 4000 mg/kg bb. Dari penelitian ini
dapat disimpulkan bahwa ekstrak etanol daun lidah mertua (EEDLM) tidak
menunjukkan gejala toksik pada kelompok kontrol dan kelompok perlakuan dosis
250, 500 dan 1000 mg/kg bb, tetapi pada kelompok perlakuan dengan dosis 2000
dan 4000 mg/kg bb ditemukan adanya gejala toksik yang ditandai dengan adanya
penurunan aktivitas motorik mencit dan aktivitas berjalan dengan perut.

ACUTE TOXICITY TEST OF ETHANOLIC EXTRACT OF MOTHER-INLAW’S TONGUE LEAVES (Sansevieria trifasciata Prain) ON MALE MICE
ABSTRACT
Generally mother-in-law’s tongue leaves (Sansevieria trifasciata Prain)

are known and cultivate as ornamental plants. Besides that, this plant can be use
as antidiabetic, antibacterial, analgesic, antipyretic, and antioxidant. The aim of
this study is to determine the potential of acute toxicity of ethanolic extract of
mother-in-law’s tongue leaves.
This experiment was designed as an experimental study by using 30 male
mice which are divided into 1 control group and 5 treatment groups, each groups
consist of 5 mice. The control group was given a suspension of CMC-Na 0.5%,
while the treatment groups were given a suspension of 250; 500; 1000; 2000 and
4000 mg/kg bw of ethanolic extract of mother-in-law’s tongue leaves. The
suspension was given once and orally at the first of the study. Intensive
observations were done in the first 4 hours to the toxic symptoms and the number
of mice that died during the 14 days and histopathological studies in livers and
kidneys. The data then were analyzed with one way ANOVA and continued with
Post Hoc test, which are LSD and Tukey.
The toxicity test showed mice that were given ethanolic extract of motherin-law’s tongue leaves in the control and the treatment group of 250; 500 and
1000 mg/kg bw did not show any toxic symptoms, but the treatment groups with
2000 and 4000 mg/kg bw showed a toxic symptom on the test stage and writhing
test but there was no dead mice during the treatment. From the observation, 4000
mg/kg bw ethanolic extract of mother-in-law’s tongue influence the SGPT value
(p < 0.05). Besides that, there were damage / necrosis liver cells in the treatment

groups of 500; 1000; 2000 and 4000 mg/kg bw. From this experiment can be
concluded that ethanolic extract of mother-in-law’s tongue leaves (EEDLM)
doesn’t show any toxic symptoms in control group and treatment groups of 250;
500 and 1000 mg/kg bw, but there is toxic symptom in treatment groups of 2000
and 4000 mg/kg bw that is pointed by the decreased motoric activity and writhing
in mice.
