Pembuatan Preparat Histologi Hati Organ Hati dicuci dengan NaCl 0,9% difiksasi (dimasukkan ke dalam fiksatif bouin) washing (dengan menggunakan alkohol 70%, dilakukan berkali- kali hingga jernih), direndam selama 1 malam dehidrasi dilakukan dengan merendam hati ke dalam alkohol bertingkat yaitu konsentrasi 30, 40, 50, 60, 70, 80, 96 dan 100% selama 1 jam pada masing-masing konsentrasi clearing dilakukan dengan merendam hati ke dalam xylol selama 1 malam Infiltrasi Embedding dilakukan dengan meletakkan hati pada kotak berbentuk segi empat yang telah dipersiapkan sebelumnya sebagai cetakan. Dituang parafin yang telah cair ke dalam kotak tersebut, kemudian hati ditanam dan diatur posisinya lalu diberi label. Dibiarkan sampai dingin dan membeku sehingga membentuk blok parafin dan dimasukkan ke dalam freezer. Dilakukan penempelan blok-blok parafin pada holder yang terbuat dari kayu berukuran 1x1 cm yang berbentuk persegi.
Blok Parafin
Cutting dilakukan dengan memotong blok-blok parafin yang telah diholder pada mikrotom sehingga membentuk pita-pita parafin dengan ukuran ketebalan 6-10 µm Attaching dilakukan dengan mengambil beberapa pita parafin dengan skapel, kemudian diletakkan pada object glass, dan dicelupkan pada air dingin dan air hangat. Lalu diletakkan di atas hotplate beberapa detik untuk melekatkan pita parafin pada object glass

Pewarnaan sediaan hati diwarnai dengan menggunakan hematoxilin Erlich selama 3-5 menit, lalu dicuci dengan dengan air mengalir ± 3 menit, kemudian dimasukkan ke dalam alkohol 30, 50, dan 70%, lalu dimasukkan ke dalam larutan pewarna Eosin 0,5% dalam alkohol selama 1-3 menit, lalu dimasukkan ke dalam aquadest dan kemudian preparat dimasukkkan berturut-turut ke dalam alkohol 30, 40, 50, 60, 70, 80, dan 96%, alkohol absolut, dikeringkan dengan kertas tisu, selanjutnya preparat dimasukkan dalam chamber berisi xylol Mounting dilakukan dengan menutup preparat dengan Canada balsam. 2. Pepaya Bangkok Deskripsi Morfologi: Varietas pepaya Bangkok dikenal juga dengan nama pepaya Thailand. Kulit luar kasar dan tidak rata. Pepaya Bangkok berukuran lebih besar dan bentuknya lebih bulat dibandingkan pepaya Cibinong. Daging buah bewarna merah jingga dan berat buah kurang lebih 3,5 kg (Kalie, 1996).

3. Pepaya Cibinong Deskripsi Morfologi: Bentuk buah panjang besar dan lancip pada bagian ujungnya. Tangkai buah panjang. Daging buah bewarna merah kekuningan, rasanya kurang manis. Berat varietas pepaya kurang lebih 2,5 kg/buah (Kalie, 1996).
c. Pepaya Cibinong
4. Pepaya Solo Deskripsi Morfologi: Ukuran buah kecil dan bentuknya mirip buah alpukat. Berat buah antara 0,4-0,1 kg/buah. Daging buah bewarna kuning, beraroma dan rasanya manis (Kalie, 1996).

5. Pepaya Burung Deskripsi Morfologi: Daging buah bewarna kuning dan berair banyak. Kulit buah bewarna hijau kekuning. Rasa asam manis (Kalie, 1996).

6. Pepaya Jinggo Deskripsi Morfologi: Daging buah bewarna merah dan berair banyak. Kulit buah bewarna kuning. Berat buah kurang lebih 1,5 kg/buah (Kalie, 1996).
7. Pepaya California Deskripsi Morfologi: Warnanya lebih mengkilap, bentuknya agak lonjong, daging buahnya tebal, bijinya lebih sedikit, dan rasanya manis (Kalie, 1996).
