Efek Pemberian Ekstrak Etanol Daun Sirsak (Annona Muricata L.) Sebagai Imunostimulan Terhadap Mencit Jantan Yang Diinduksi Dengan Prednison

muricata L.) SEBAGAI IMUNOSTIMULAN PADA MENCIT
JANTAN YANG DIINDUKSI DENGAN PREDNISON
ABSTRAK
Imunostimulan adalah senyawa yang dapat meningkatkan mekanisme
pertahanan tubuh baik secara spesifik maupun non spesifik. Senyawa yang
ditunjukan untuk perbaikan fungsi imun pada kondisi- kondisi imunosupresi.
Pertahanan non spesifik terhadap antigen ini disebut paramunitas, dan zat
berhubungan dengan penginduksi disebut paraimunitas. Induktor semacam ini
biasanya tidak atau sedikit sekali kerja antigennya, akan tetapi sebagian besar
bekerja sebagai mitogen yaitu meningkatkan poliferasi sel yang berperan pada
imunitas. Sel-sel yang mengalami imunostimulasi adalah makrofag, granulosit,
limfosit T dan B karena induktor ini bekerja menstimulasi mekanisme pertahanan
seluler.
Daun sirsak (Annona muricata L.), famili Annonaceae merupakan salah
satu tumbuhan yang diduga memiliki aktivitas imunostimulan.
Tujuan dari penelitian ini adalah untuk melihat aktivitas imunostimulan
ekstrak etanol daun sirsak pada mencit jantan yang diinduksi dengan suspensi
prednison 0,05%.
Pada penelitian uji aktivitas imunostimulan suspensi ekstrak etanol daun
sirsak jumlah total sel darah putih dan diferensiasi sel darah putih mencit jantan
yang diinduksi dengan suspensi prednison 0,05% dosis 5 mg/kg BB secara oral.
Uji aktivitas imunostimulan suspensi ekstrak etanol daun sirsak diberikan secara
oral dosis 15, 25 dan 35 mg/kgBB, 1x setiap hari, dimulai hari kedua selama 7
hari, sebagai pembanding diberikan Stimuno® dengan dosis 25 mg/kgBB.
Dari hasil penelitian didapat bahwa ekstrak daun sirsak pada dosis 35
mg/kg BB menunjukkan kemampuan dalam meningkatkan jumlah sel darah putih
dengan nilai 9,4 x 103 ± 0,6772. Untuk jumlah limfosit pada dosis 35 mg/kg BB
menunjukkan nilai 8,6 x 103 ± 0,6593. Untuk jumlah mid sel (monosit, eosinofil, basofil,
sel blast dan prekursor sel darah putih) pada dosis 35 mg/kg BB menunjukkan nilai 0,6 x
103 ± 0,0632. Untuk jumlah sel granulosit pada dosis 35 mg/kg BB menunjukkan nilai
0,216 x 103. Dari hasil pengujian statistik, pemberian ekstrak etanol daun sirsak
mampu meningkatkan jumlah total sel darah putih dan diferensial sel darah putih
secara signifikan terhadap kelompok uji (p < 0,05).

EFFECT OF SOURSOP LEAF (Annona muricata L.) ETHANOL
EXTRACT AS TO THE IMUNOSTIMULATORY IN
PREDNISON INDUCED MALE MICE

Immunostimulatory are certain compounds that can increase the body's
defense mechanisms, both specific and non-specific. Coumpunds shown to
improve immune function in immunosuppressed conditions. Non specific defence
against this antigen are called paraimunitas, and substance associated with know
inducers. Inductor is usually no or very little antigenic work, but most of the work
as a mitogen that increases proliferation of cells involved in immunity.
Destination cell is macrofage, granulocytes, lymphocytes T and B because the
inductor's work stimulates cellular defense mechanisms.
Soursop leaves (Annona muricata L.), family Annonaceae is one plant that
is thought to have immunostimulatory activity.
The purpose of this study is to look at the immunostimulatory activity of
soursop leaf extract in prednisone 0,05% induced male mice.
In this research, the immunostimulatory activity test of soursop leaf extract
was done to the count of white blood cells and white blood cell differentiation
parameter of prednisone induced male mice with the dose of prednisone 5 mg/kg
body weight (BW). The soursop leaf ethanol extract was given orally with the
dose of 15, 25 and 35 mg/kg BW 1x per day, starting the second day for seven
days, Stimuno® as to the comparison was given orally with the dose 25 mg/kg
BW.
From the results obtained that soursop leaf extract at a dose of 35 mg/kg
BW showed the efficacy to increase the amount of white blood cells with a value
of 9.4 x 103 ± 0.6772. For lymphocyte counts at doses of 35 mg/kg showed the
value of 8.6 x 103 ± 0.6593. To the number of mid cells (monocytes, eosinophils,
basophils, blast and other precursor white cells) dose of 35 mg/kg BW showed the
highest increase in value with the value of 0.6 x 103 ± 0,0632. To the number of
granulocyt dose of 35 mg/kg BW showed the highest increase in value with the
value of 0.216 x 103. Based on the result of statistical test, the administration of
soursop leaf ethanol extract was able to increase the count of white blood cells
and white blood cell differentiation value significantly against the test group (p <
0.05).
