In this research, it can be shown that the treatment of gabapentin 100 mg in a day twice during 1 month had significant effect in decreasing the rate of pain, this is
appropriate to the theory mentioning that gabapentin could be felt the effect after 1-2 week of treatment and help to reduce the pain if it was used at least 2-3
months Acute Pain Service, 2010.
8. The Therapeutic Effect of Amitriptilin to Reduce Pain
Amitriptilin is the medicine from tricyclic antidepressant group TCA with the mechanism action to hamper the up-take 5-HT and norepineprin NE; besides
this also decrease the receptor 5-HT, so it can increase the concentration 5-HT in sinaptik gap. The hamper of re-uptake NE also increased the concentration of NE
in sinaptik gap. The increase of NE caused decrease of receptor adrenergik beta number that reduced adenilsiklasi activities which was automatically decrease
adenosum monofosfat and decrease the opening of sinaps-Na. the decrease of Sinaps-Na which opened means that depolarization decreased and the pain also
decreased Richeimir S, 2003. The treatment of amitriptilin caused decrease rate 1.278 from the week before giving the treatment until the week 2 after giving the
treatmnet. Significant value from the range 2 weeks got 0.000 p 0.005 showing decrease 1.278 that was different statistically. The treatment of amitriptilin after 4
weeks mathematically consisted decrease 2.444. From the statistic test, the difference of VAS value in the week 0 and the week 4 amitriptilin group was
achieved significance value 0.000 p 0.005 showing the reduction 2.87 which was statistically different. This study is able to show that the treatment of
amitriptilin 12.5 mg in a day twice during 1 month had significant effect in reducing the rate of pain in the patients.
