Lampiran 1 Prosedur ekstraksi karaginan Gliksman 1983 yang dimodifikasi
Rumput laut kering Kappaphycuss alvarezii sebanyak 100 - 500 gram dicuci bersih sampai semua kotoran yang menempel seperti karang, pasir dan garam
hilang. Untuk mempercepat proses ekstraksi ukuran rumput laut dikecilkan dengan cara di-blender. Ekstraksi dilakukan dengan menggunakan air pada suhu

C dengan perbandingan rumput laut kering dengan air 1 : 40 selama 3 jam. Selama proses ekstraksi pH dipertahankan sekitar 8,5–9,0 dengan cara
menambahkan NaOH 1. Proses pemisahan selulosa prefiltrasi dilakukan dengan menggunakan
nylon mesh ukuran 350 mesh. Proses tersebut dilanjutkan kembali dengan menggunakan filter ukuran 1 atau 0,3
μ. Sistem yang penyaringan yang digunakan pada proses ini adalah sistem dead end.
Proses pemurnian dan pengkonsentrasian karaginan dilakukan dengan menggunakan membran mikrofiltrasi 0,1
μ sistem cros-flow. Rententat dari proses mikrofiltrasi diendapkan dengan isopropil alkohol dan atau langsung
dikeringkan sehingga diperoleh refined carrageenan kering.
